摘要
探究Aurora B在宫颈癌组织中的表达水平以及抑制Aurora B对宫颈癌细胞增殖、侵袭及迁移能力的影响,采集未经放疗和化疗的宫颈癌组织和子宫肌瘤组织(对照),提取总RNA,实时荧光定量PCR(RFQ-PCR)检测Aurora B在宫颈癌组织中的表达水平,分析Aurora B与宫颈癌发生的相关性。以不同量的Aurora B抑制剂AZD1152-HQPA处理SiHa、HeLa和293FT(对照)细胞,CCK-8细胞增殖检测法检测对细胞增殖的抑制率,Transwell小室侵袭和Transwell迁移实验检测细胞侵袭和迁移能力。Aurora B mRNA在宫颈癌中的表达(0.00388±0.000560)极显著高于(p<0.01)在子宫肌瘤中的表达量(0.00103±0.000161),表达上调3.76倍。AZD1152-HQPA抑制Aurora B激酶活性时,宫颈癌细胞系实验组的侵袭细胞数和迁移细胞数均极显著低于对照组(p<0.01);且AZD1152-HQPA浓度从50 nmoL升高至100 nmoL时,侵袭细胞数和迁移细胞数极显著降低(p<0.01)。Aurora B在宫颈癌组织中表达水平显著提高,其可能通过促进癌细胞增殖、侵袭和迁移来促进肿瘤的发生,是宫颈癌诊断和治疗的潜在靶标之一。
To investigate the expression level of Aurora B in cervical cancer tissues and the inhibitory effect of Aurora B on the proliferation,invasion and migration of cervical cancer cells.cervical cancer tissues and uterine fibroid tissues(control)without radiotherapy and chemotherapy were collected(control),tissues total RNA was extracted,the expression level of Aurora B in cervical cancer tissues was detected by RFQ-PCR,and the correlation between Aurora B and occurrence of cervical cancer was analyzed.SiHa,HeLa and 293 FT cells(control)were treated with different doses of Aurora B inhibitor AZD1152-HQPA.The inhibition rate of cell proliferation were analyzed by CCK-8 cell proliferation assay.Transwell invasion assay and Transwell migration assay were used to detect cell invasion and migration ability.the expression of Aurora B mRNA in cervical cancer(0.00388±0.000560)was obviously higher(p<0.01)than that in uterine fibroids(0.00103±0.000161)and the expression was increased by 3.76 fold.When AZD1152-HQPA inhibited Aurora B kinase activity,the number of invasive cells and migratory cells in cervical cancer cell lines were decreased obviously than those in control group(p<0.01);and the concentration of AZD1152-HQPA was increased from 50 nmoL to 100 nmoL,The number of invasive cells and the number of migrated cells significantly decreased(p<0.01).The expression level of Aurora B in cervical cancer tissue is significantly increased,which may promote the occurrence of tumor by promoting the proliferation,invasion and migration of cancer cells,and is a potential target for the diagnosis and treatment of cervical cancer.
