配伍延胡索对马钱子毒性影响的实验研究 被引量:6

Experimental study on the toxicity of compatibility of Rhizoma Corydalis on Semen Strychni

摘要 目的:研究配伍延胡索对马钱子毒性的影响,为马钱子临床合理、安全用药提供科学依据。方法:选择马钱子生品和延胡索醋炙品为药材,采用急性毒性实验测定马钱子及马钱子与不同延胡索配比水煎液的LD50值;应用高效液相色谱法测定各种水煎液中士的宁和马钱子碱的含量;应用电喷雾质谱法对马钱子与延胡索配伍后主要生物碱的变化进行研究。结果:与延胡索配伍后,配伍药对的毒性随着延胡索比例的增加而增加;马钱子等量配伍延胡索后,药对煎液中士的宁和马钱子碱的溶出率与马钱子生品相比分别下降了46.27%和57.58%,随着延胡索用量的增加,士的宁和马钱子碱的含量继续下降。结论:延胡索可以增强马钱子的毒性,药对煎液的毒性与有毒生物碱的含量并不成正比关系,马钱子与延胡索合煎后,延胡索中生物碱成分抑制了马钱生物碱的溶出。药对煎液的毒性可能与延胡索成分的煎出有关,或与合煎后产生了毒性成分有关。 Objective:To study the toxicity of Rhizoma Corydalis to Semen Strychni,and to provide scientific basis for clinical safe application of Semen Strychni.Methods:The crude drugs of Semen Strychni and Rhizoma Corydalis were selected as medicinal materials,LD50 values of Semen Strychni and decoction with different ratio of Rhizoma Corydalis to Semen Strychni were determined by acute toxicity test.The contents of strychnine and brucine in water decoction were determined by HPLC;the changes of major alkaloids in Rhizoma Corydalis and Semen Strychni were studied by electrospray mass spectrometry(ESI MS).Results:After compatibility with Rhizoma Corydalis,the toxicity of compatibility drug pairs increased with the increase of the proportion of Rhizoma Corydalis.The dissolution rates of strychnine and brucine in the decoction after the same amount of Semen Strychni and Rhizoma Corydalis were decreased respectively by 46.27% and 57.58% compared with the crude drug.With the increase of Rhizoma Corydalis dosage,strychnine and brucine contents continued to decrease.Conclusion:Rhizoma Corydalis can incresase the toxicity of Semen Strychni,the toxicity of medicine to decoction is not proportional to the content of toxic alkaloids.After Semen Strychni and Rhizoma Corydalis tuber are decocted together,alkaloids in Rhizoma Corydalis inhibit the dissolution of brucine alkaloids.The toxicity of the medicine to the decoction may be related to the decoction of Rhizoma Corydalis,or to the generation of toxic components after the decoction.
