摘要
为研究胍基乙酸中药制剂的毒性作用,进行了胍基乙酸中药制剂的急性毒性试验研究。小鼠在试验环境中适应1 d后,按体重随机分为对照组和不同剂量的试验组(500、1000、2000、4000和8000 mg/kg),每组10只(雌雄各半),给药开始前禁食不禁水12 h,1次灌服给药,给药4 h后正常饲喂。对照组给等量生理盐水,给药后连续观察4 h,其余时间每天观察3次小鼠精神状况,连续观察7 d。对小鼠进食状况、饮水情况、有无出现异常活动、有无异常肌肉运动、对外界刺激的反应等毒性反应情况和死亡情况进行详细记录。试验观察结束后,处死动物并解剖,对小鼠的脏器进行观察。结果表明:试验期间小鼠精神状况正常,无小鼠死亡。对照组与不同剂量试验组的脏器各项指数变化不明显(P>0.05)。与对照组相比,各试验组AST指标极显著降低(P<0.01);试验2、3、4组ALT相对于对照组均极显著降低(P<0.01),试验1、5组ALT低于对照组但差异不显著(P>0.05);各试验组ALP均低于对照组(P<0.01)。试验2、3、5组与对照组相比,T-AOC指标差异极显著(P<0.01);试验1、2、3组与对照组相比,GSH-P X极显著增加(P<0.01);试验2、3、4组与对照组相比,CAT极显著增加(P<0.01),同样与试验1、5组相比,极显著增加(P<0.05);试验2、3组与对照组相比,MDA极显著降低(P<0.01);各试验组SOD与对照组相比,均差异极显著(P<0.01)。综上所述,胍基乙酸中药制剂无毒性,安全,具有抗氧化、保护肝脏的作用。
In order to study the toxic effect of traditional Chinese medicine preparation of glycidyl acetate,the acute toxicity test of traditional Chinese medicine preparation of guanidine acetate was carried out.The mice were randomly divided into the control group and the test groups(500 mg/kg,1000 mg/kg,2000 mg/kg,4000 mg/kg and 8000 mg/kg)according to their weight after being adapted to the test environment for 1 d,each group was consisted of 10 mice(half male and half female).The mice were fasted for 12 h before administration,and were given by gavage once,and were fed normally 4 h after administration.The control group was given normal saline.After administration,the mice were observed continuously for 4 h,and the rest of the time was observed 3 times a day.The mental state of the mice was observed continuously for 7 d.The toxic reactions such as eating,drinking water,abnormal activity,abnormal muscle movement and response to external stimulation and deaths of mice were recorded in detail.At the end of the experiment,the animals were killed and dissected to observe the organs of mice.The results showed that the mental state of mice was normal and no mice died.There was no significant change in organ indexes between the control group and the experimental group(P>0.05).Compared with the control group,the AST in each experimental group was significantly lower(P<0.01);the ALT in test 2,3,4 groups was significantly lower than that of the control group(P<0.01),while the ALT in test 1,5 groups was lower than that of the control group(P>0.05);the ALP in each experimental group was lower than that of the control group(P<0.01).Compared with the control group,the difference of T-AOC index in test 2,3 and 5 groups was significant(P<0.01);compared with the control group,GSH-Px in test 1,2,3 and 4 groups was significantly increased(P<0.01);compared with the control group,CAT in test 2,3 and 4 groups was significantly increased(P<0.01),MDA in test 2 and 3 groups was significantly decreased(P<0.01);SOD in each experimental group was significantly different from that in the control group(P<0.01).In conclusion,guanidine acetic acid Chinese medicine preparation is nontoxic,safe,and has the function of antioxidation and liver protection.
