Study on Tissue Culture of Populus adenopoda Maxim

摘要 以响叶杨叶片为外植体进行组培快繁,对外植体消毒、诱导培养、增殖培养、生根培养及炼苗移栽进行了深入研究,结果表明:0.10%升汞溶液最佳消毒时间为5.5 min;最佳诱导培养基为处理1-7(MS+1.00 mg·L-16-BA+0.10 mg·L-1 NAA),诱导率为90.0%;最佳增殖培养基为处理2-8(MS+0.40 mg·L-16-BA+0.15 mg·L-1 NAA),增殖倍数为4.0;适宜生根培养基为1/2 MS+0.10 mg·L-1 NAA,生根率为98.0%;炼苗后移栽选用树皮∶珍珠岩∶黄土=1∶1∶1的基质,成活率达92.0%。研究指出,通过芽苞嫁接、温室培育方法,有利于响叶杨野外采集的外植体脱毒。 Study on tissue culture and rapid propagation of the leaves explants of Populus adenopoda Maxim,the results showed that:the optimum disinfection time of 0.1%mercury solution was 5.5 min;the optimum induction medium was treatment 1-7(MS+1 mg·L-16-BA+0.1mg·L-1 NAA),the induction rate was 90.0%;the optimum multiplication medium was treatment 2-8(MS+0.4 mg L-16-BA+0.15 mg·L-1 NAA),the multiplication rate was 4.0;the optimal root culture medium is 1/2 ms+0.1 mg·L-1 NAA,the rooting rate was 98.0%;after seedling refining,the bark was transplanted∶perlite∶loess soil=1∶1∶1,the survival rate was 92.0%.It was pointed out that the method of bud grafting and greenhouse cultivation was beneficial to the detoxification of explants collected in the field.
