摘要
为建立洋葱愈伤诱导及再生体系,本试验以8个不同洋葱品种花梗为外植体进行愈伤组织诱导及再生苗等研究。结果表明,黑暗、25℃条件下,诱导培养基为MS+1.8 mg/L 2,4-D+2.0 mg/L甘氨酸+700mg/L脯氨酸+1.0 g/L干酪素水解物+30 g/L蔗糖+8 g/L琼脂时,不同洋葱花梗均能形成愈伤组织,诱导率为100%,表明该诱导过程不受基因型影响。不同洋葱花梗愈伤组织再生苗培养基为MS+30 g/L蔗糖+8g/L琼脂,其诱导成苗过程与生根同步,移栽成活率达90%;‘环球’的再生频率最高,为30%。
In order to establish the callus induction and regeneration system of onion,the pedicels of 8 different onion varieties were used as explants to study the callus induction and regeneration seedlings.The results showed that under the conditions of darkness and 25℃,all materials in the culture medium of MS + 1.8 mg/L 2,4-D + 2.0 mg/L glycine + 700 mg/L proline + 1.0 g/L casein hydrolysate + 30 g/L sucrose + 8 g/L agar could induce callus,and the induction rate was 100%,which indicated that the induction process was not affected by genotype.MS + 30 g/L sucrose + 8 g/L agar was the medium for regeneration seedlings of different onion pedicel calli.The process of plantlet formation was synchronous with rooting,and the survival rate of transplanting reached 90%.The regeneration frequency of ‘Huanqiu’was the highest and up to 30%.
