摘要
酪氨酸酶抑制剂在医药、化妆品、食品工业等领域应用广泛,本文旨在建立一种能用于该抑制剂研究的快速筛选方法。本研究以曲酸为阳性对照,通过紫外分光光度法检测反应体系的吸光度变化,以判断待测物质对酪氨酸酶的抑制作用。通过优化最终建立了最佳筛选方案为:最大吸收波长为475nm,底物最适浓度为2.5mg/mL酶最适浓度为0.2mg/mL。基于此模型筛选方法,对128个备用的发酵液提取物进行抑制活性的初步筛选,共筛选获得2个对酪氨酸酶具有高抑制活性的样品。结果证实本方法可以用于酪氨酸酶抑制剂的快速筛选,方法简单、易行、稳定、可靠,是寻找和发现有潜在活性的酪氨酸酶抑制剂的有效手段。
Tyrosinase inhibitors are widely used in medicine,cosmetics,food industry and other fields,so we aim to establish a rapid screening method of tyrosinase inhibitors.In this study,kojic acid was used as a positive control,and the change of absorbance of the reaction system was detected by ultraviolet spectrophotometry,so as to judge the inhibition effect of the substance to be tested on tyrosinase.The optimal screening scheme was established through optimization:the maximum absorption wavelength was 475 nm,the optimum substrate concentration was 2.5 mg/mL,and the optimum enzyme concentration was 0.2 mg/mL.Based on this model,128 fermentation broth extracts were screened for their inhibitory activities,and two samples with high inhibitory activities for tyrosinase were obtained.The results show that this method can be used to screen tyrosinase inhibitors quickly and provide a simple,easy,stable and reliable research platform.It is an effective way to find and search potential tyrosinase inhibitors.
