母系表达基因3对卵巢癌细胞侵袭和迁移能力的影响机制研究

摘要 目的探究长链非编码RNA母系表达基因3(maternally expressed gene 3,MEG3)对卵巢癌细胞基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)和基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)的表达和侵袭、迁移能力的影响。方法荧光定量聚合酶链反应检测卵巢上皮细胞和卵巢癌细胞MEG3表达情况。通过慢病毒转染携带MEG3全长序列的载体和空白载体于SKOV3人卵巢癌细胞中,分别作为实验组细胞和对照组细胞,通过Transwell侵袭实验检测两组细胞的侵袭能力,通过Transwell迁移实验检测两组细胞的迁移能力,通过蛋白质印迹法检测两组细胞MMP-2和MMP-9的表达情况。结果人卵巢癌细胞SKOV3、OVCAR3和A2780中MEG3的表达显著低于人卵巢上皮细胞IOSE80(均P<0.05)。通过慢病毒转染外源性上调SKOV3细胞MEG3的表达,实验组细胞MEG3表达显著高于对照组(P<0.05)。Transwell侵袭实验结果显示实验组穿过基质胶的细胞数显著少于对照组(P<0.05);Transwell迁移实验结果显示实验组迁入下室的细胞数显著少于对照组(P<0.05)。蛋白质印迹法结果显示实验组细胞MMP-2和MMP-9表达水平均显著低于对照组(均P<0.05)。结论MEG3低表达于人卵巢癌细胞系,并可下调MMP-2和MMP-9的表达,抑制卵巢癌细胞的侵袭和迁移能力。 Objective To investigate the effects of long non-coding RNA maternally expressed 3(MEG3)on the expression of matrix metalloproteinase-2(MMP-2)and matrix metalloproteinase-9(MMP-9)and the ability of invasion and migration of ovarian cancer cells.Method Fluorescence quantitative polymerase chain reaction(PCR)technique was used to detect the expression of MEG3 in ovarian epithelial cell and ovarian cancer cells.The lentiviral vector and empty vector were transfected with the full-length MEG3 sequence of SKOV3 in human ovarian cancer cell line,and were respectively regarded as experimental group and control group.Transwell invasion assay was used to detect the invasive ability of the two groups cells.Transwell migration was used to detect the migration ability of the two groups cells.Western Blotting was used to detect the expression of MMP-2 and MMP-9.Result The expression level of MEG3 in ovarian cancer cells including SKOV3,OVCAR3 and A2780 were significantly lower than that in ovarian epithelial cell IOSE80(all P<0.05).The expression of MEG3 in SKOV3 cells was up-regulated by lentivirus transfection,and the expression level of MEG3 in experimental group was significantly higher than that in control group(P<0.05).Transwell invasion assay showed that the number of cell in experimental group through the matrigel glue was significantly less than that in control group(P<0.05).Transwell migration assay showed that the number of cell in experimental group through the chamber was significantly less than that in control group(P<0.05).Western Blotting showed that the expression levels of MMP-2 and MMP-9 in experimental group were significantly lower than those in control group(all P<0.05).Conclusion MEG3 is low expressed in human ovarian cancer cell,and can inhibit cell invasion and migration by down-regulating the expression of MMP-2 and MMP-9.
