高铜诱导肉鸡肌胃和腺胃发生自噬与凋亡 被引量:5

High levels of copper induce autophagy and apoptosis in muscular stomach and glandular stomach of broiler chickens

摘要 为了探讨高铜对肉鸡肌胃和腺胃发生自噬与凋亡的影响,本试验选用240羽1日龄AA白羽肉鸡作为试验动物,并随机分为4组,分别饲喂含铜11 mg/kg(对照组)、110 mg/kg(高铜Ⅰ组)、220 mg/kg(高铜Ⅱ组)和330 mg/kg(高铜Ⅲ组)的日粮,并于49日龄采集肌胃和腺胃,制作组织切片观察组织学变化,采用实时荧光定量PCR检测自噬和凋亡相关基因的mRNA表达,免疫印迹检测LC3-Ⅱ/LC3-Ⅰ和Caspase3的蛋白表达。结果显示,高铜可引起肌胃中肌纤维稀疏,腺胃中的腺细胞发生脱落以及空泡变性。另外,高铜可显著上调肌胃和腺胃中自噬相关基因Beclin1、Atg5、LC3-Ⅰ、LC3-Ⅱ和凋亡相关基因Bax、Bak1、p53、Cyt C和Caspase3的表达,LC3-Ⅱ/LC3-Ⅰ和Caspase3的蛋白表达水平也显著升高,而mTOR和Bcl2的mRNA转录水平则显著下调。此外,腺胃和肌胃中p62的mRNA转录水平呈现先降低后升高的趋势。结果表明,高铜可诱导肉鸡的肌胃和腺胃发生自噬和凋亡。 To investigate the effects of high levels of copper on autophagy and apoptosis in muscular stomach and glandular stomach of broiler chickens,a total of 240 one-day-old AA white feather broiler chicks were randomly divided into 4 groups in this study.The broilers were fed the diet with 11 mg/kg copper(control group),110 mg/kg copper(high-copper groupⅠ),220 mg/kg copper(high-copper groupⅡ),and 330 mg/kg copper(high-copper groupⅢ),respectively.The muscular stomach and glandular stomach were collected on day 49 and tissue section was made for histological observation.The mRNA expression of autophagy and apoptosis-related genes were detected by real-time fluorescent quantitative PCR and the protein expression levels of LC3-Ⅱ/LC3-Ⅰand Caspase3 were determined by Western-blot.The results showed that high levels of copper could cause muscle fiber sparsity in muscular stomach and defluvium and vacuolar degeneration in gland cells of glandular stomach.Additionally,the mRNA expression of autophagy-related genes(Beclin1,Atg5,LC3-Ⅰ,and LC3-Ⅱ),apoptosis-related genes(Bax,Bak1,p53,Cyt C,and Caspase3),and protein expression of LC3-Ⅱ/LC3-Ⅰand Caspase3 in muscular stomach and glandular stomach were significantly elevated with increasing levels of copper.However,the mRNA expression levels of mTOR and Bcl2 were decreased remarkably with the increased copper.Furthermore,the mRNA level of p62 in muscular stomach and glandular stomach was decreased firstly and then increased.The results suggested that high levels of copper could induce autophagy and apoptosis in muscular stomach and glandular stomach of broiler chickens.
