马铃薯低温胁迫的生理响应及耐寒性综合评价 被引量:13

Physiological Responds of Potato Seedings to Low Temperature Stress and Comprehensive Evaluation on Their Cold Tolerance

摘要 【目的】为探究低温胁迫下不同生理指标对耐寒性评价的影响。【方法】以16份马铃薯盆栽苗为材料,离体叶片置于程序降温仪进行低温胁迫,测定植物叶片过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、过氧化物酶(POD)活性和游离脯氨酸(PRO)、可溶性糖(SS)、可溶性蛋白(SP)、丙二醛(MDA)含量。采用主成分分析、隶属函数和聚类分析方法综合评价供试材料的耐寒性。【结果】耐寒性强弱排序依次为:‘黄麻子’>‘AV1’>‘F170’>‘AV9’>‘华彩1号’>‘陇薯8号’>‘acanle-4’>‘中薯11号’>‘396034.268’>‘300046.22’>‘中薯8号’>‘冀张14号’>‘陇薯7号’>‘acanle-6’>‘E1’>‘634777’。【结论】综合分析可知,低温胁迫下马铃薯叶片中可溶性蛋白含量、丙二醛含量和超氧化物歧化酶活性可作为评价马铃薯耐寒性的重要参考指标。 【Objective】This paper aimed to explore the effect of different physiological indexes on cold tolerance evaluation under low temperature stress.【Method】With 16 potted potato seedlings as materials,in vitro leaves were subjected to low temperature stress by a program temperature-reducing apparatus to determine the activities of catalase(CAT)active,superoxide dismutase(SOD)active,peroxidase(POD)and content of free proline(PRO),soluble sugar(SS),soluble protein(SP)and malondialdehyde(MDA)in plant leaves.Principal component analysis,membership function and cluster analysis were used to evaluate the cold tolerance of the tested materials.【Result】The order of cold tolerance was as follows:‘Huangmazi’>‘AV1’>‘F170’>‘AV9’>‘Huacai No.1’>‘Longshu No.8’>‘acanle-4’>‘Zhongshu No.11’>‘396034.268’>‘300046.22’>‘Zhongshu No.8’>‘Jizhang No.14’>‘Longshu No.7’>‘acanle-6’>‘E1’>‘634777’.【Conclusion】The comprehensive analysis showed that soluble protein content,malondialdehyde content and SOD activity in potato leaves under low temperature stress were used as important reference indicators for evaluating potato cold tolerance.
