摘要
目的建立检测抗-D浓度的竞争酶免疫分析法(EIA)检测抗-D浓度,探讨抗-D浓度与效价的相关性。方法应用EIA法检测抗-D标准品浓度并建立标准曲线,收集18(人)份抗-D阳性血浆并使用EIA法检测其抗-D浓度;同时用试管法检测18份标本的抗-D效价。采用简单线性回归分析抗-D浓度与效价的相关性。结果抗-D标准品浓度(1.78—28.5)IU/mL在EIA法检测标准曲线的线形范围内,r2>0.99;抗-D浓度(3.2—43.7)IU/mL时,抗-D效价为4—256,二者的回归模型Y=6.829+0.158X(X为效价,Y为浓度),回归方程r2=0.932(P<0.01)。结论EIA法可定量检测抗-D浓度,抗-D浓度与效价呈良好的相关性。
Objective To investigate the correlation between anti-D and titer by detecting the concentration of anti-D using competitive enzyme immunoassay.Methods EIA was used to detect the concentration of anti-D standard substance and establish the standard curve.18 anti-D positive plasma was collected and the anti-D concentration was detected by EIA.The anti-D titer of 18 samples was measured by test tube method.The correlation between anti-D concentration and anti-D titer was analyzed by simple linear regression.Results The standard curves were generated using serial dilutions of anti-D,and significant correlation was found between the EIA and anti-D concentration(IU/mL)in the range 1.78 to 28.5,as the r2 value approached more than 0.99.The regression model of anti-D concentration and titer was Y=6.829+0.158X(X was titer and Y was concentration),and the r2 of the regression equal to 0.932(P<0.001).Conclusion EIA can quantitatively detect the concentration of anti-D,and the concentration of anti-D has a good correlation with titer.
