沉默Paralemmin-3表达对缺氧复氧致心肌细胞损伤的保护作用

摘要 目的探讨沉默Paralemmin-3(PALM3)表达对缺氧复氧致心肌细胞损伤的保护作用。方法将H9c2细胞随机分为4组,即对照组、模型组、siRNA-Control组、siRNA-PALM3组。对照组使用正常细胞,模型组使用缺氧复氧损伤的细胞;siRNA-Control组和siRNA-PALM3组分别使用siRNA-Control和siRNA-PALM3质粒转染的细胞,转染48 h后进行缺氧复氧损伤。比较4组细胞间的生存率;检测上清液中白介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)的含量;检测细胞中PALM3、核因子kappa B(NF-κB)和单免疫球蛋白白介素-1受体相关蛋白(SIGIRR)的表达。结果与对照组相比较,模型组细胞中PALM3 mRNA和蛋白质的表达量均升高(P<0.05)。与模型组和siRNA-Control组相比较,siRNA-PALM3组细胞中PALM3 mRNA和蛋白质的表达量均降低,存活率升高,上清液中IL-1β和TNF-α的含量均降低,NF-κB蛋白质的表达量降低,SIGIRR蛋白质的表达量升高(P<0.05)。结论在缺氧复氧损伤中,PALM3的表达显著升高,使用RNA干扰沉默PALM3的表达,可以提高细胞的存活率,降低炎性因子的分泌和NF-κB的表达,可能与促进SIGIRR的表达有关。 Objective To investigate the protective effects of silencing Paralemmin-3(PALM3)expression on hypoxia/reoxygenation-induced cardiomyocytes injury in vitro.Methods H9c2 cells were randomly divided into four groups:control group,model group,siRNA-control group and siRNA-PALM3 group,and the normal cells were used as control group,hypoxia/reoxygenation-induced cells were used as model group,and the cells in siRNA-control group and the siRNA-PALM3 group were transfected with siRNA-control and siRNA-PALM3 plasmids,respectively,which were subjected to hypoxia/reoxygenation injury for 48h after transfection.The cell survival rates were observed and compared among the four groups.The levels of interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)in the supernatant were detected.Moreover the expression levels of PALM3,nuclear factor kappa B(NF-κB)and single immunoglobulin IL-1 receptor-related molecule(SIGIRR)in the cells were detected.Results As compared with those in control group,the expression levels of PALM3 mRNA and protein in model group were significantly increased(P<0.05).As compared with those in model group and the siRNA-control group,the expression levels of PALM3 mRNA and protein in siRNA-PALM3 group were significantly decreased.In addition the survival rate was increased,the contents of IL-1βand TNF-αin the supernatant were significantly decreased,the expression levels of NF-κB protein were significantly decreased,however,the expression levels of SIGIRR protein were significantly increased(P<0.05).Conclusion During the hypoxia/reoxygenation injury,the expression levels of PALM3 are significantly increased.Silencing PALM3 by RNA interference can increase cell survival rate,inhibit the secretion of inflammatory factors and decrease the expression levels of NF-κB,which may be related to the increase of SIGIRR expression.
