摘要
目的研究人参对阿霉素所致心肌细胞损伤的治疗机制。方法筛选ADR损伤心肌的最佳时间和剂量、研究人参对阿霉素(ADR)损伤心肌细胞乳酸脱氢酶(LDH)漏出量、自噬和凋亡相关基因的表达量、搏动频率的影响。结果 (1)心肌细胞损伤条件为浓度8.00 mg/L的阿霉素(ADR)作用心肌细胞12h。(2)人参能提高受损心肌细胞的搏动频率。(3)人参可以使损伤心肌细胞LDH的漏出量显著降低。(4)人参对阿霉素(ADR)损伤心肌细胞Caspase-3的m RNA表达量具有抑制作用,有显著性差异;对Beclin-1的m RNA表达效果强于人参组(P<0.001);而对Atg5的m RNA的表达的影响只有人参组体现出差异(P<0.01)。结论人参可以对阿霉素(ADR)所致损伤的心肌细胞乳酸脱氢酶(LDH)释放量产生抑制作用、提高阿霉素(ADR)所致损伤的心肌细胞搏动频率、抑制阿霉素(ADR)所致损伤的心肌细胞凋亡及自噬相关基因m RNA的表达量,说明人参对阿霉素(ADR)所致损伤的心肌细胞具有保护作用。
Objective To study the therapeutic mechanism of ginseng on adriamycin-induced cardiomyocyte injury. Methods The optimal time and dose of ADR-injured myocardium were screened. The effects of ginseng on the expression of lactate dehydrogenase (IDH), the expression of autophagy and apoptosis-related genes and the frequency of pulsation in adriamycin (ADR). Results ① Cardiomyocytes were treated with adfiamycin (ADR) at the concentration of 8.00 mg I L for 12 h. ② Ginseng can increase the frequency of impaired cardiomyocytes. ③ Ginseng can significantly reduce the leakage of LDH in cardiomyocytes. (P〈0.001). The expression of Beclin-1 mRNA was significantly higher than that of Ginseng (P〈0.001). The expression of Caspese-3 mRNA in adriamycin (ADR) The effect of mRNA expression was only on the ginseng group (P〈0.01). Conclusion Ginseng can inhibit the release of lactate dehydrogenase (LDH) in cardiomyocytes induced by adriamycin (ADR), increase the frequency of cardiomyocyte pulsation induced by adriamycin (ADR), inhibit the (ADR) induced damage of myocardial cells and autophagy-related gene mRNA expression, indicating that ginseng on the dmnage caused by adriamycin (ADR) cardiomyocytes have a protective effect.
