不同剂量当归对硫化钠致小鼠斑秃模型的影响 被引量:6

Effects of Different Doses of Angelica Sinensis on Mice Model of Alopecia Areata Induced by Sodium Sulfide

摘要 目的目的:探讨当归对斑秃小鼠血清细胞间黏附分子-1(ICAM-1)、内皮细胞-白细胞黏附分子-1(ELAM-1)的影响。方法将40只雄性KM小鼠随机分为空白对照组(Control),模型组(Model),当归低、中、高(Low、Medium、High dose)剂量组。除空白对照组外,均以8%硫化钠酒精溶液局部外涂,制作斑秃模型。分别以0.9%生理盐水和当归水煎剂对空白对照组、模型组、当归低、中、高剂量组给药,每天1次,连续7 d。采用酶联免疫吸附测定(ELISA)和苏木-伊红素(HE)染色法分别检测小鼠血清中细胞间黏附分子-1(ICAM-1)、内皮细胞-白细胞黏附分子-1(ELAM-1)水平和计算单位视野毛囊数;同时对实验小鼠背部脱毛区毛发生长情况进行评分。结果①当归各剂量组与模型组比较,均可提高斑秃小鼠脱毛区毛发评分和单位视野毛囊数(P<0.05);②当归各剂量组与模型组比较,均可降低血清ICAM-1和ELAM-1水平(P<0.05);③当归不同剂量组两两比较中,中剂量组生发效果最好(P<0.05)。结论当归对硫化钠致斑秃小鼠模型有生发作用,且以中剂量效果最佳。其生发作用可能通过降低血清ICAM-1和ELAM-1水平,进而抑制毛囊的细胞凋亡和毛囊的的退化,促进毛发生长。 Objective To investigate the effects of Danggui(Angelica sinensis)on serum intercellular adhesion molecule-1(ICAM-1)and endothelial cell-leukocyte adhesion molecule-1(ELAM-1)in alopecia areata mice.Methods Forty male KM mice were randomly divided into control group(Control),model group(Model),Danggui(Angelica sinensis)low,medium and high dose groups(Low,Medium,High dose).Alopecia areata model was prepared by topical application of 8%sodium sulfide alcohol solution except the blank control group.The rats in the blank control group,the model group and the low,medium and high dose groups were given Danggui(Angelica sinensis)with 0.9%normal saline and Danggui(Angelica sinensis)water decoction,respectively,once a day for 7 days.Enzyme-linked immunosorbent assay(ELISA)and hematoxylin-eusin(HE)staining were used to detect the serum levels of intercellular adhesion molecule-1(ICAM-1)and endothelial cell-leukocyte adhesion molecule-1(ELAM-1)in mice and calculate the number of hair follicles per unit of visual field.At the same time,the hair growth of the back hair removal area of the experimental mice was scored.Results(1)The scores of hair loss and the number of hair follicles per unit of visual field in alopecia areata of Danggui(Angelica sinensis)groups were improved compared with those of the model group(P<0.05).(2)The serum levels of ICAM-1 and ELAM-1 in Danggui(Angelica sinensis)groups can be reduced compared with those of the model group(P<0.05).(3)Danggui(Angelica sinensis)middle dose group had the best hair growth effect compared with the high and low dose groups(P<0.05).Conclusion Danggui(Angelica sinensis)has a hair growth effect on mice model of alopecia areata and the effect of the middle dose is the best.Its hair growth effect may promote hair growth by reducing serum levels of ICAM-1 and ELAM-1,thereby inhibiting hair follicle apoptosis and hair follicle degradation.
