一种硒代蛋氨酸衍生物的合成表征与安全性评价

摘要 以L-硒代蛋氨酸、月桂酰氯和油酰氯为主要原料通过肖顿-鲍曼缩合和酯化反应得到了N-酰基-L-硒代蛋氨酸衍生物,产物运用核磁共振(NMR)技术(1H NMR,13 C NMR)表征确认。采用噻唑蓝(MTT)比色法研究了目标化合物对人体正常肝细胞(HL-7702)和大鼠肾小管上皮细胞(NRK-52E)的细胞毒性。结果表明,合成的目标化合物对人体正常肝细胞(HL-7702)和大鼠肾小管上皮细胞(NRK-52E)细胞毒性较低,当2种目标化合物的浓度达1920μmol/L时,细胞存活率均大于75%,证明合成的硒代蛋氨酸衍生物是一种低毒的硒代化合物,对于扩大硒代蛋氨酸在油脂类食品和农产品加工领域的应用具有一定潜力。 N-acyl-L-selenomethionine derivatives were synthesized by Schotten-Baumann condensation using L-selenomethionine,lauroyl chloride and oleoyl chloride as the main raw materials,and then esterified with methanol to obtain the N-acyl-L-selenomethionine methyl ester.The product was characterized and confirmed by nuclear magnetic resonance(NMR)technology(1H NMR,13 C NMR).In addition,the cytotoxicity of the target compound on human normal liver cells(HL-7702)and rat renal tubular epithelial cells(NRK-52E)was studied by MTT colorimetry.The results showed that two synthesized target compounds were less cytotoxic to human normal liver cells(HL-7702)and rat renal tubular epithelial cells(NRK-52E).When the concentration of two target compounds reached 1920μmol/L,the cell survival rate was greater than 75%,which proved that the synthetic selenomethionine derivative is a low-toxic selenomethine compound and has certain potential for expanding selenomethionine in the field of oily food and agricultural products processing.
