微小RNA-599在膀胱癌组织的表达及其临床意义 被引量:7

Expression and significance of microRNA-599 in bladder cancer

摘要 目的检测膀胱癌患者组织和正常膀胱组织中微小RNA(miRNA,miR)-599的相对表达水平,及转染miR-599后对膀胱癌5637细胞增殖、迁移和侵袭能力的影响。方法收集2016年1月至2018年6月间泉州第一医院的40例膀胱癌患者的标本和40例正常膀胱组织标本,使用实时定量反转录聚合酶链反应(RT-qPCR)法检测miR-599的相对表达水平,分析其与患者临床病理因素之间的相关性。实验分为miR-599模拟体(mimics)组和阴性对照组(NC)。使用细胞计数试剂盒(CCK-8)法、划痕实验和侵袭小室(Transwell)观察过表达miR-599后,对5637细胞的增殖、迁移和侵袭能力的影响。组间比较采用两独立样本t检验。患者临床病理特征的分析采用χ^2检验。结果与正常膀胱组织比较(1.068±0.451),膀胱癌组织miR-599相对表达水平(0.362±0.290,χ^2=14.040,P<0.05)明显下调,且与患者与肿瘤TNM分期明显相关(χ^2=4.177,P<0.05)。膀胱癌5637细胞系的miR-599相对表达量(0.313±0.087)明显低于正常膀胱上皮细胞,差异有统计学意义(1.096±0.164,P<0.05)。与NC组比较,5637细胞的相对增殖率为(51.2±4.5)%、迁移率为(32.4±6.2)%和侵袭率为(39.2±7.3)%,比例均明显下降,差异有统计学意义(P<0.05)。结论miR-599在膀胱癌组织中的表达呈低表达状态,且与膀胱癌患者肿瘤分期(TNM分期)明显相关。miR-599的表达水平与膀胱癌细胞的增殖、迁移和侵袭有关,过表达miR-599能够明显抑制5637细胞的增殖、迁移和侵袭能力。 Objective To explore the expression of microRNA(miRNA,miR)-599 in bladder cancer and to detect the effects of miR-599 overexpression on proliferation,migration and invasion of 5637 bladder cancer cell line.Methods Real-time quantitative reverse transcriptase-polymerase chain reaction(RT-qPCR)was used to detect the expression of miR-599 in 40 cases of bladder cancer tissues and normal bladder specimens.We then analyzed the relationship between the expression of miR-599 and the clinicopathological data of bladder cancer patients.After transfection with miR-599,the effects on proliferation,migration and invasion were evaluated using cell counting kit-8(CCK-8)assay,wound healing assay and Transwell assay.Results The relative expression of miR-599 was significantly lower in bladder cancer tissues(0.362±0.290,P<0.05)than in normal bladder tissues(1.068±0.451).The level of miR-599 was positively correlated with patients’TNM stage(TNM,P<0.05).The expression of miR-599 in 5637 cell line(0.313±0.087)were significantly lower than those in normal bladder epithelial cell line(1.096±0.164,P<0.05).The proliferation[(51.224±4.506)%,P<0.01],migration[(32.436±6.204)%,P<0.05]an invasion[(39.257±7.331)%,P<0.01]of 5637 cells with miR-599 mimic transfection were significantly lower than those in miR-599 negative control after transfection.Conclusion The expression of miR-599 was decreased in patients with bladder cancer.The low level of miR-599 was significantly associated with the primary tumor/regional lymph nodes/distant metastasis(TNM)stage.It was shown that miR-599 was associated with proliferation,migration and invasion of 5637 cells.The overexpression of miR-599 could suppress the capability of proliferation,migration and invasion.
