美味牛肝菌不同极性溶剂萃取物的抗氧化及抑菌活性研究 被引量:2

Antioxidant and Antibacterial Properties of Extracts from Different Polar Solvents from Boletus edulis Bull.

摘要 研究美味牛肝菌不同极性溶剂萃取物的抗氧化及抑菌效果,为其活性成分的开发利用提供依据。用不同极性溶剂对美味牛肝菌乙醇浸膏进行分级萃取,得到石油醚相、乙酸乙酯相、正丁醇相和水相萃取物4部分,评价4种萃取物清除DPPH自由基(DPPH·)、ABTS+自由基(ABTS+·)、羟自由基(·OH)和超氧阴离子自由基(·O2-)能力;以滤纸片琼脂扩散法和微孔板2倍稀释法测定4种萃取物对3种革兰氏阴性菌(大肠杆菌、铜绿假单胞菌、变形杆菌)和3种革兰氏阳性菌(金黄色葡萄球菌金黄亚种、枯草芽孢杆菌、四联球菌)的抑菌活性。结果表明,美味牛肝菌不同极性溶剂萃取物对·OH清除力高于Vc,清除能力依次为石油醚相(EC50为(0.016±0.002) mg/mL)>乙酸乙酯相(EC50为(0.191±0.007) mg/mL)>水相(EC50为(0.207±0.007) mg/mL)>正丁醇相(EC50为(0.225±0.007) mg/mL),4种萃取物对·O2-清除力相当(EC50均为0.007 mg/mL),对DPPH·和ABTS+·有一定的清除能力但低于Vc。美味牛肝菌4种萃取物对6种菌株都有一定程度的抑制效果,其中乙酸乙酯相和水相对四联球菌的抑菌效果最好,最低抑菌浓度均为12.5 mg/mL。美味牛肝菌中含有较多抗氧化和抑菌的活性物质,其中乙酸乙酯相的抗氧化活性和抑菌活性相对较高,可作为今后分离美味牛肝菌中活性成分的研究重点。 In this study,the antioxidant activity and antibacterial activity of four different solvent extracts from Boletus edulis Bull.were investigated.It could be provided a basis for the further development and utilization of bioactive compounds of Boletus edulis Bull..The ethanol extract was by successive extraction with petroleum ether,ethyl acetate,n-butyl alcohol,and distilled water.The DPPH· scavenging ability,ABTS+· scavenging ability,·OH scavenging ability and ·O2-scavenging ability of the four extracts were determined,and all the extracts were evaluated for their bacteriostatic effect using agar diffusion and broth microdilution method,and three gram-negative bacteria (Escherichia coli,Pseudomonas aeruginosa and Proteus vulgaris) and three gram-positive bacteria (Staphylococcus aureus subsp.aureus,Bacillus subtilis and Micrococcus tetragenus) were determined in our research.The results showed that the scavenging activity of the four extracts from Boletus edulis Bull.on ·OH was higher than that of Vc,and the antioxidant activities of petroleum ether extract (EC50=(0.016±0.002) mg/mL) was the strongest,followed by ethyl acetate extract (EC50=(0.191±0.007) mg/mL),aqueous extract (EC50=(0.207±0.007) mg/mL),n-butyl alcohol extract (EC50=(0.225±0.007) mg/mL).The four extracts had the same scavenging activities on ·O2-(EC50=0.007 mg/mL) as Vc,and which had a certain scavenging activity on DPPH· and ABTS+· but less than Vc.The antibacterial activities of the four extract phases of Boletus edulis Bull.were effective to 6 kinds of bacterias.Ethyl acetate extract and aqueous extract showed the most antibacterial activities,and the minimum bactericidal concentration against Micrococcus tetragenus was 12.5 mg/mL.Boletus edulis Bull.contains abundant antioxidant and antibacterial substances.In conclusion,ethyl acetate extract had better antioxidant and antibacterial activities,and it can be used as the research emphasis of the further study on the isolation of the active ingredients from Boletus edulis Bull..
