摘要 为建立全红杨组织培养与快繁体系,以全红杨叶柄为外植体,进行不同激素配比对愈伤组织诱导、不定芽分化、生根培养等影响研究。结果表明:适宜愈伤组织诱导培养基为MS+6-BA0. 2 mg·L^-1+NAA 0. 05 mg·L^-1+蔗糖25. 0 g·L^-1+琼脂6. 0 g·L^-1,诱导率96. 67%;适宜不定芽分化培养基为MS+6-BA 0. 5 mg·L^-1+NAA 0. 2 mg·L^-1+蔗糖30. 0 g·L^-1+琼脂6. 0 g·L^-1,分化率100%;适宜生根培养基为1/2MS+IBA 0. 3 mg·L^-1+蔗糖20. 0 g·L^-1+琼脂6. 0 g·L^-1,生根率100%。本研究也为选育杨树良种、种质资源保存、扩大栽培与推广等提供研究基础。 In order to establish the tissue culture and fast propagation system of Populus × euramericana cv.‘Quanhong’,the effects of different ratio of hormones on callus induction,adventitious bud differentiation and rooting culture were studied by using the leaf stalk as explant. The results showed that suitable medium for callus induction was MS +6-BA 0. 2 mg·L^-1+ NAA 0. 05 mg·L^-1+ sucrose 25. 0 g·L^-1+ agar 6. 0 g·L^-1,with the induction rate 100%;suitable medium for adventitious bud differentiation was MS + 6-BA 0. 5 mg·L^-1+ NAA 0. 2 mg·L^-1+ sucrose 30. 0 g·L^-1+ agar6. 0 g·L^-1,with the differentiation rate 100%;suitable medium for rooting was 1/2 MS + IBA 0. 3 mg·L^-1+ sucrose 20. 0 g·L^-1+ agar 6. 0 g·L^-1,with rooting rate 100%. This study also provides the research foundation for the poplar breeding,the germplasm resource preservation,the expansion of cultivation and promotion.
