摘要
目的建立检测中药饮片中黄曲霉毒素的QuEChERS净化样品结合超高效液相色谱-串联质谱法。方法色谱柱采用Agilent Eclipse XDB C18 RRHD柱(50 mm×2.1 mm,1.8μm),流动相为2 mmoL/L乙酸铵溶液(含0.1%甲酸)-乙腈(梯度洗脱),采用电喷雾离子源模式(ESI)、动态多反应监测(DMRM)正离子扫描下采集数据。结果4种黄曲霉毒素在相应测定范围内线性关系良好,相关系数均大于0.99,在低、中、高不同浓度加标水平下,平均回收率为88.47%~108.06%(n=6)。结论该方法简便快速、选择性好、灵敏度高,适用于中药饮片中黄曲霉毒素的快速检测。
Objective To establish a method for the detection of aflatoxins in traditional Chinese medicines prepared in ready-to-use forms with QuEChERS purification by ultra performance liquid chromatography-triple quadrupole mass spectrometry(UPLC-MS/MS).Methods The samples were separated on an Agilent Eclipse XDB C18 RRHD column(50 mm×2.1 mm,1.8μm).The mobile phase composed of 2 mmoL/L ammonium acetate solution(containing 0.1% formic acid)and acetonitrile with gradient elution.Dynamic multi-reaction monitoring(DMRM)scanning was employed for quantification with switching electrospray ion(ESI)source polarity in positive ions.Results The four kinds of aflatoxins showed good linear relationships within their own ranges,the correlation coefficients were above 0.99.Under different concentration levels of low,medium and high,their average recoveries ranged from 88.47% to108.06%(n=6).Conclusion The method is simple,rapid,highly selective and sensitive,which can be used for the rapid analyzing of aflatoxins in traditional Chinese medicines prepared in ready-to-use forms.
