番茄红素对脓毒症大鼠免疫及炎性因子的影响 被引量:2

摘要 目的探讨番茄红素对脓毒症大鼠免疫及炎性细胞因子的影响。方法将30只SD大鼠随机分为对照组、模型组、处理组,每组10只。处理组大鼠以番茄红素预处理2 h后腹腔注射内毒素,模型组大鼠腹腔注射内毒素制备脓毒症模型后给予等量生理盐水,对照组大鼠给予等量生理盐水。6 h后采血,检测各组大鼠外周血中CD3^+,CD8^+,CD4^+T细胞百分比及白细胞介素6(IL-6)、肿瘤坏死因子-α(TNF-α)、丙二醛(MDA)、超氧化物歧化酶(SOD)水平,采血后将其处死,并取肺组织行苏木精-伊红(HE)染色。结果与对照组相比,模型组大鼠外周血中CD4^+T细胞百分比及IL-6,TNF-α,MDA水平均显著升高(P<0.05),CD3^+,CD8^+T细胞百分比及SOD水平显著降低(P<0.05);与模型组相比,处理组大鼠外周血中CD4^+T细胞百分比及IL-6,TNF-α,MDA水平均显著降低(P<0.05),CD3^+,CD8^+T细胞百分比及SOD水平均显著升高(P<0.05);与对照组相比,处理组大鼠外周血中CD3^+,CD8^+,CD4^+T细胞百分比及SOD,MDA,IL-6,TNF-α水平均有显著差异(P<0.05);HE染色显示,处理组大鼠肺泡结构、肺泡间隔、肺间质充血水肿、中性粒细胞浸润较模型组均有明显改善。结论番茄红素能有效降低脓毒症大鼠氧化应激反应,抑制炎性细胞因子产生,减轻肺组织损伤,但对脓毒症发病机体免疫功能异常改变的作用不明显。 Objective To investigate the effect of lycopene on immunity and inflammatory cytokines in sepsis rats.Methods A total of30 SD rats were randomly divided into the control group,model group and treatment group,10 rats in each group.The treatment group was pretreated with lycopene for 2 h,and then injected with endotoxin.The model group was injected with endotoxin to prepare a sepsis model,and then injected with equal amount of normal saline.The control group was given the same amount of normal saline.After 6 h,the percentage of CD3^+,CD8^+,CD4^+T cells and interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),malondialdehyde(MDA)and superoxide dismutase(SOD)levels in peripheral blood of each group were detected.Pathological changes of lung tissue in each group were observed by Hematoxylin Eosin(HE)staining after execution.Results Compared with the control group,the percentage of CD4+T cells and IL-6,TNF-α,and MDA levels in the peripheral blood of the model group were significantly increased(P<0.05),and the percentages of CD3^+,CD8^+T cell and the level of SOD were significantly reduced(P<0.05);compared with the model group,the percentage of CD4^+T cells and IL-6,TNF-α,MDA levels in the peripheral blood of the treatment group were significantly reduced(P<0.05),and the percentages of CD3^+,CD8^+T cells and the level of SOD were significantly increased(P<0.05);compared with the control group,the percentages of CD3^+,CD8^+CD4^+T cells and the level of SOD,MDA,IL-6,and TNF-αwere significantly different(P<0.05);HE staining showed that the alveolar structure,alveolar interval,pulmonary interstitial congestion and edema,and neutrophil infiltration in the treatment group had significantly improved compared with the model group.Conclusion Lycopene can effectively reduce the oxidative stress response of septic rats,inhibit the production of inflammatory cytokines,and alleviate lung tissue damage;but it has no obvious effect on the abnormal changes of immune function in the body with sepsis.
