摘要
目的:将HPD100与D101大孔吸附树脂纯化禾本科芦笋总黄酮与总酚酸,优选总黄酮与总酚酸纯化工艺。方法:测定静态吸附率、解吸率、静态吸附曲线来评价HPD100大孔吸附树脂与D101大孔树脂对芦笋中总黄酮与总酚酸的静态吸附性能;然后对径高比、洗脱溶剂、水洗体积、洗脱剂量等考察获取动态纯化工艺。结果:总黄酮与总酚酸在HPD100大孔树脂上静态吸附量为分别为38.23 mg·g-1与22.40 mg·g-1,解吸率大小分别为68.95%与79.05%。动态优选的结果为浓度1∶0.25,上样量4 BV,径高比1∶4,静置4 h,9 BV水洗,50 ml 90%乙醇洗脱,总黄酮与总酚酸纯度分别达34.90%与20.07%。总黄酮与总酚酸在D101大孔树脂上静态吸附量为分别为40.05 mg·g-1与32 mg·g-1,解吸率大小分别为62.78%与71.74%。动态优选的结果为浓度1∶0.25,上样量3 BV,径高比1∶5,静置4 h,7 BV水洗,60 ml 90%乙醇洗脱,总黄酮与总酚酸纯度分别达34.10%与17.27%。结论:HPD100与D101大孔吸附树脂可以用于禾本科芦笋中总黄酮与总酚酸有效部位的纯化。
Objective:To obtain the purification process of total flavonoids and phenolic acids in Asparagus from Gramineae by studying the purification of total flavonoids and phenolic acids by HPD100 and D101 macroporous resins.Methods:The static adsorption rate,desorption rate and static adsorption curve were determined to evaluate the static adsorption performances of HPD100 and D101macroporous resins for total flavonoids and total phenolic acid in Asparagus,and then the dynamic purification process was obtained by investigating the diameter-height ratio,elution solvent,washing volume and elution dose.Results:The static adsorption capacity of total flavonoids and phenolic acids by HPD100 macroporous resin was 38.23 mg·g-1 and 22.40 mg·g-1,respectively.The desorption rate was 68.95%and 79.05%,respectively.The results of dynamic optimization were as follows:4 BV sample with the concentration of 1∶0.25,the diameter-height ratio of 1∶4,and standing for 4 hours,and then washed by 9 BV and 50 ml 90%ethanol.The purity of total flavonoids and phenolic acids was 34.90%and 20.07%,respectively.The static adsorption capacity of total flavonoids and phenolic acids by D101 macroporous resin was 40.05 mg·g-1 and 32 mg·g-1,respectively.The desorption rate was 62.78%and 71.74%,respectively.The results of dynamic optimization were as follows:3 BV sample with the concentration of 1∶0.25,the diameter-height ratio of 1∶5,and standing for 4 hours,and then washed by 7 BV and 60 ml 90%ethanol.The purity of total flavonoids and phenolic acids was 34.10%and 17.27%,respectively.Conclusion:The total flavonoids and phenolic acids in Asparagus from Gramineae can be purified by HPD100 and D101 macroporous resins with the optimized process.
