摘要
试验采用反胶束体系进行己酸乙酯的猪胰脂肪酶催化合成,并用气相色谱检测.己酸乙酯最佳转化条件为:温度为35℃,酶质量浓度为0.003 g/L,己酸浓度为0.3 mol/L,醇酸比为1∶1.气相色谱检测采用外标法定量.结果表明,己酸乙酯色谱纯含量在0.1~1.0μL范围内呈良好线性关系,相关系数为0.9909;己酸乙酯的回收率在87.47%~95.76%;相对标准偏差RSD为2.98%.说明该方法能够准确对反胶束体系脂肪酶催化合成的己酸乙酯进行定性和定量检测.
Ethyl caproate was synthesized from caproic acid and anhydrous ethanol catalyzed by porcine pancreas lipase in reversed micellar systems. The optimal reaction conditions: reaction temperature 35 ℃;enzyme concentration 0.003 g/L;ratio of ethanol to acid 1:1;caproic acid concentration 0.3 mol/L. External standard quantitative method was used for gas chromatography.The results showed that the linear scopes were between 0.1 μL and 1.0 μL(R=0.9859);The recovery rate of GC Detection was between 87.47% and 95.76% with the Relative Standard Deviation is 2.98%. The method can be simple, fast and accurate to detect ethyl caproate contents synthesized in reversed micellar systems.
