摘要
建立超临界流体色谱法控制3-乙酰基邻苯二甲酸酐(阿普斯特中间体)。用亚乙基桥杂化颗粒为填充剂(WatersTorus1-AA,3.0×100mm,1.7μm);以CO2-0.05%三氟乙酸乙腈溶液(35︰65)为流动相,等度洗脱;流速为每分钟1.5 mL;检测波长为220 nm;补偿吸光度为360~400nm;柱温为55℃;进样盘温度为5℃;ABPR压力为2000 psi。采用超临界流体色谱技术,以超高效合相色谱系统能够完全分离3-乙酰氨基邻苯二甲酸酐与3-乙酰氨基邻苯二甲酸,能有效控制3-乙酰氨基邻苯二甲酸酐,是一种环保、简便、高效的分析方法。
A supercritical fluid chromatography method was established to control 3-acetyl phthalic anhydride(the key intermediate of Apremilast). Ethylene bridge hybrid particles as filler(Waters Torus 1-AA, 3.0×100 mm, 1.7 μm);CO2-0.05 % trifluoroacetic acid acetonitrile solution(36︰64) as mobile phase, isocratic elution;The flow rate is 1.5 mL per minute;the detection wavelength is 220 nm;the compensated absorbance is 360~400 nm;the column temperature is 55 ℃;the sample tray temperature is 5 ℃;the ABPR pressure is 2000 psi. In this study, the use of supercritical fluid chromatography technology and ultra-high performance combined chromatography system can completely separate 3-acetaminophthalic anhydride and 3-acetaminophthalic acid, meanwhile, effectively control 3-acetaminophthalic anhydride. It is an environmentally friendly, simple and efficient analysis method.
