摘要
目的探讨石军方免煎颗粒剂(SJF)对急性肝衰竭小鼠肝组织的影响。方法雌性C57BL/6J小鼠75只随机分为五组。6只作为空白对照组。模型对照组(24只)和给药组(45只)腹腔注射对乙酰氨基酚600 mg/kg建立急性肝衰竭小鼠模型。给药组均分为三组,造模前3 d分别行SJF溶液0.2 mL/20 g(低剂量组)、0.4 mL/20 g(中剂量组)或0.8 mL/20 g(高剂量组)灌胃,每天1次,连续3 d。模型对照组和空白对照组用等量生理盐水灌胃。造模24 h后处死小鼠,检测肝功能,HE染色观察肝组织病理变化,免疫组化检测肝组织Toll样受体4(TLR4)蛋白表达水平。结果与空白对照组比较,模型对照组小鼠DBil、ALT、AST升高而白蛋白降低,肝/脾重量比、肝组织坏死评分和TLR4蛋白表达水平升高(P<0.05或P<0.01)。与模型对照组比较,三个给药组小鼠DBil和TLR4蛋白表达水平降低,中、高剂量组ALT降低,低、高剂量组AST和肝/脾重量比降低,高剂量组白蛋白升高,低剂量组小鼠肝组织坏死评分降低(P<0.05或P<0.01)。结论SJF能减少急性肝衰竭小鼠的肝细胞坏死,改善肝功能及精神状态,降低死亡率;其减轻肝损伤的作用机制可能是通过抑制肝组织TLR4蛋白的表达。
Objective To explore the effect of Shijunfang granules(SJF)on liver tissues of the mice with acute liver failure.Methods Seventy-five female C57 BL/6 J mice were randomly assigned into 5 groups.Six mice were taken as blank controls(group BC).The mouse models with acute hepatic failure were established by intraperitoneal injection of acetaminophen 600 mg/kg in group MC(model control,24 mices)and three SJF-treated groups(45 mice).Before acetaminophen injection,liquid SJF was given by gastric gavage in the doses of 0.2 mL/20 g(group LS,15 mice),0.4 mL/20 g(group MS,15 mice)or 0.8 mL/20 g(group HS,15 mice)once a day for three days.The same amount of normal saline instead of SJF was used in groups of BC and MC.The mice of 5 groups were executed after 24 hours.Liver function was detected,the pathological changes of liver tissues were observed,and the expression of Toll-like receptor 4(TLR4)protein was detected by immunohistochemistry.Results Compared to group BC,the mice of group MC showed higher levels of DBil,ALT and AST,lower albumin,and higher weight ratio of liver to spleen,necrosis score of liver tissues and expression of TLR4 protein(P<0.05 or P<0.01).Compared with group MC,the mice of groups of MS and HS showed lower ALT,the mice of groups of LS and HS showed lower AST and weight ratio of liver to spleen,the mice of group HS showed higher albumin,the mice of group LS showed lower necrosis score of liver tissues(P<0.05 or P<0.01).Conclusion SJF can reduce the necrosis of liver cells,improve the liver function and mental status,reduce the mortality in the mice with acute liver failure.The mechanism for SJF to reduce the liver damage may be its inhibitory effect on TLR4 protein expression in liver tissues.
