SIRT1/TLR4/NF-κB在丹参素治疗大鼠心肌梗死的保护作用 被引量:13

摘要 目的探讨SIRT1/TLR4/NF-κB在丹参素治疗大鼠心肌梗死的保护作用。方法构建SD大鼠心肌缺血再灌注损伤模型,随机分为:对照组,模型组、丹参素组(2 mg/kg)三组,每组15只。HE染色法观察心肌组织病理学改变;TTC双染法测定心肌梗死面积;免疫组织化学法检测SIRT1表达;采用Western blot法检测SIRT1、TLR4、NF-κB、Bcl-2、Caspase3蛋白的表达;ELISA法检测心肌组织IL-6和TNF-α表达。结果丹参素组大鼠心肌凋亡数量、心肌梗死面积比模型组显著降低(P<0.05)。丹参素组SIRT1表达平均光密度显著升高(P<0.05)。丹参素组TLR4、NF-κB、Caspase3蛋白表达明显下降,SIRT1、Bcl-2蛋白含量表达比模型组显著上升(P<0.05);丹参素组心肌组织IL-6和TNF-α表达显著下降(P<0.05)。结论丹参素可通过促进SIRT1表达,抑制TLR4/NF-κB信号通路来改善大鼠心肌梗死,对心肌起到营养和保护的作用。 Objective To discuss the protective effect of silent information regulator 1(SIRT1),target lesion revascularization rate 4(TLR4)and nuclear factor-κB(NF-κB)in rats with myocardial infarction treated with Danshensu(DSS).Methods The model of myocardial ischemia-reperfusion injury was established in SD rats,and then the rats were divided randomly into control group,model group and DSS group(2 mg/kg,each n=15).The pathological changes of myocardial tissue were observed after HE staining,and size of myocardial infarction was detected after TTC double staining.The expression of SIRT1 was detected by using immunohistochemistry technique.The protein expressions of SIRT1,TLR4,NF-κB,Bcl-2 and Caspase3 were detected by using Western blotting assay.The expressions of myocardial interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)were detected by using ELISA.Results The number of cardiomyocyte apoptosis and size of myocardial infarction decreased significantly in DSS group compared with model group(P<0.05).The mean optical density of SIRT1 increased significantly in DSS group(P<0.05).The protein expressions of TLR4,NF-κB and Caspase3 decreased significantly,and protein expressions of SIRT1 and Bcl-2 increased significantly in DSS group compared with control group(P<0.05).The expressions of myocardial IL-6 and TNF-αdecreased significantly in DSS group(P<0.05).Conclusion DSS can relieve myocardial infarction in rats and plays a role in nutrition and protection of myocardium through promoting SIRT1 expression and inhibiting TLR4/NF-κB signaling pathway.
