摘要
目的:探讨积雪草颗粒在肾缺血再灌注损伤中的保护作用。方法:健康SPF级雄性SD大鼠30只,分为3组,每组10只,分别为假手术组、模型组、积雪草颗粒组。假手术组在造模前40 d及造模后3天用等量生理盐水灌胃(10 mL·kg^-1),每日1次,造模时仅暴露双侧肾蒂45 min,不予以特殊处理,然后逐层缝合关腹。模型组灌胃剂量与假手术组相同,造模时采用无创动脉夹夹闭双侧肾蒂45 min,然后松开无创血管夹让肾脏血流复灌,然后逐层缝合关腹。积雪草颗粒组在造模前40 d及造模后3 d用积雪草颗粒水溶液灌胃(折合成生药5 g·kg^-1,10 mL·kg^-1),每日1次,其余操作同模型组操作。结果:①肾功能指标:模型组血肌酐[(38.1000±5.8013)μmol·L^-1]、尿素氮[(9.7600±1.6008)mmol·L^-1]水平明显高于假手术组[(24.8000±3.4254)μmol·L^-1,(5.5700±0.8206)mmol·L^-1](P<0.05),积雪草颗粒组血肌酐、尿素氮水平[(31.9000±4.7714)μmol·L^-1,(7.6300±1.6600)mmol·L^-1]明显低于模型组(P<0.05),但高于假手术组(P<0.05)。②肾脏病理切片评分:模型组[(2.9800±0.5203)分]明显高于假手术组[(0.3000±0.3300)分,P<0.05],积雪草颗粒组[(1.5200±0.3011)分,P<0.05]高于假手术组(P<0.05),但低于模型组(P<0.05)。③细胞凋亡水平:模型组细胞凋亡水平(0.7761±0.1067)明显高于假手术组(0.1591±0.0414),积雪草颗粒组细胞凋亡水平(0.4611±0.0741)高于假手术组(P<0.05),但低于模型组(P<0.05)。结论:积雪草颗粒在肾缺血再灌注损伤中发挥重要作用,可减轻血液再灌注后肾脏细胞的凋亡、保护肾功能。
Objective:To explore the protective effect of Jixuecao Granule on renal ischemia-reperfusion injury.Methods:30 healthy SPF male SD rats were divided into three groups,10 in each group,which were sham operation group,model group and Jixuecao Granule group.In the sham operation group,the rats were perfused with the same amount of normal saline(10 mL·kg^-1)40 days before and 3 days after modeling,once a day,and only exposed bilateral renal pedicles for 45 minutes,without special treatment,and then closed the abdomen layer by layer.The dose of gavage in model group was the same as that in sham operation group.The renal pedicle was closed by noninvasive artery clamp for 45 minutes,then the noninvasive artery clamp was released to allow the renal blood flow to be reperfused,and then the abdomen was closed by suture layer by layer.In the group of Jixuecao Granule,the water solution of Jixuecao Granule was infused into stomach 40 days before and 3 days after modeling(converted into 5 g·kg^-1 and 10 mL·kg^-1 of crude drug),once a day,and the other operations were the same as those in the model group.Results:①Renal function indexes:the levels of creatinine[(38.1000±5.8013)μmol·L^-1],urea nitrogen[(9.7600±1.6008)mmol·L^-1]in model group were significantly higher than those in sham operation group[(24.8000±3.4254)μmol·L^-1,(5.5700±0.8206)mmol·L^-1](P<0.05),and the levels of creatinine and urea nitrogen[(31.9000±4.7714)μmol·L^-1,(7.6300±1.6600)mmol·L-1]in Jixuecao Granule group was significantly lower than the model group(P<0.05),but higher than the sham operation group(P<0.05).②The score of renal pathological section in the model group[(2.9800±0.5203)score]was significantly higher than that in the sham operation group[(0.3000±0.3300)score,P<0.05],and that in the Jixuecao Granule group[(1.5200±0.3011)score,P<0.05]was higher than that in the sham operation group(P<0.05),but lower than that in the model group(P<0.05).③Apoptosis level:the apoptosis level of model group(0.7761±0.1067)was significantly higher than that of sham operation group(0.1591±0.0414),and that of Jixuecao Granule group(0.4611±0.07414)was higher than that of sham operation group(P<0.05),but lower than that of model group(P<0.05).Conclusion:Jixuecao Granule plays an important role in renal ischemia-reperfusion injury,which can reduce renal cell apoptosis and protect renal function after blood reperfusion.
