槟榔不同炮制品对斑马鱼急性毒性比较研究 被引量:4

摘要 目的:通过比较槟榔不同炮制品对斑马鱼的LC50值及其生物碱、鞣质类成分的含量,初步探讨槟榔的毒性成分。方法:采用高效液相色谱法、鞣质含量测定法分别对槟榔各炮制品水提物中的生物碱、鞣质类成分进行含量测定,并用其不同浓度的水提物培养发育正常的斑马鱼幼鱼24h,统计斑马鱼死亡情况,采用Bliss法计算LC50值。结果:槟榔不同炮制品水提物中总生物碱的含量依次为:生槟榔>炒槟榔>焦槟榔,其总鞣质含量依次为:炒槟榔>生槟榔>焦槟榔;生槟榔、炒槟榔和焦槟榔对斑马鱼幼鱼的LC50值分别为29.74μg/mL、32.47μg/mL和40.03μg/mL;其中槟榔各炮制品水提物中生物碱的含量与其LC50值呈负相关,而总鞣质的含量与LC50值不具有相关性。结论:炒槟榔、焦槟榔对斑马鱼的急性毒性较生槟榔低,符合中药炮制减毒的基本原理;推测生物碱类成分可能是槟榔的毒性成分,其鞣质类成分的毒性有待于进一步研究。 Objective:To explore the toxicity of arecae semen by comparation the acute toxicity of different processed arecae semen on zebrafish.Methods:The alkaloids and tannins of processed arecae semen were tested by HPLC and tannin measuring process,then Zebrafish were exposed to its aqueous extraction with different concentration for 24hand their LC50were calculated,respectively.Results:The concentration of alkaloids in different processed arecae semen were raw arecae semen>fried arecae semen>coked arecae semen,and the tannins in different processed arecae semen were fried arecae semen>raw arecae semen>coked arecae semen.The LC50value of raw,fried and coked arecae semen were 29.74μg/mL,32.47μg/mL and 40.03μg/mL,respectively.Conclusion:The toxicity of the raw areca semen is much stronger than that of fried and coked arecae semen which is coincident with the toxicityreducing effect of processing.The alkaloids are the poisonous component of arecae semen and the toxicity of tannins still need further research.
