摘要
在垂直潜流人工湿地中,微生物分泌胞外物质,导致孔隙率减小。结合胞外物质在影响活性污泥性能和人工湿地除堵方面的研究,依据垂直流人工湿地反应器基质胞外多糖提取物的结构分析,选取β-葡聚糖酶,研究其对胞外多糖提取物的酶解效果。得出理论条件下其最佳酶解条件:底物质量浓度为2.8 mg/L,酶质量浓度为1.2 mg/L,体系pH值为5.5,温度为50℃,酶解时间为10 min。效果影响的主次顺序为:底物浓度>酶质量浓度>温度>pH值。
In vertical-flow constructed wetland,microbial propagation can secrete extracellular substances,resulting in the decrease of porosity.In response to its impact on activated sludge performance and the artificial wetland,and based on the structural analysis of extracellular polysaccharide extract from the substrate of the vertical-flow constructed wetland reactor,β-glucanase was selected to study its enzymolysis effect on the extracellular polysaccharide extract.The results showed that the optimum enzymolysis conditions of experimental wetland reactor matrix extracellular polysaccharide extract:substrate was 2.8 mg/L,enzymatic concentration was 1.2 mg/L,pH value was 5.5,temperature was 50℃and reaction time was 10 min.The results indicated that the sequence order effect of the four factors was:substrate concentration>enzyme concentration>temperature>pH.
