摘要
目的:观察蒙药忠伦阿汤对胶原诱导性关节炎(CIA)模型大鼠滑膜细胞凋亡相关因子的影响。方法:通过牛Ⅱ型胶原建立CIA大鼠模型,将造模成功后SD大鼠随机分为模型组,忠伦阿汤高、中、低剂量组,甲氨蝶呤组,灌胃6周,观察给药前后大鼠体质量、右后足跖厚度及关节炎指数的变化,光镜下观察踝关节病理学改变,免疫组织化学法检测滑膜组织中Bax、Bcl-2、caspase-3的表达。结果:忠伦阿汤可降低大鼠足跖厚度及关节炎指数,减轻踝关节病理学改变;与模型组比较,忠伦阿汤高、中剂量组可使Bax、caspase-3表达显著升高(P<0.01),Bcl-2表达显著降低(P<0.01)。结论:蒙药忠伦阿汤可能通过调节凋亡相关因子的表达,诱导滑膜细胞凋亡,减少关节损伤。
Objective:To observe the effects of Mongolian medicine Zhonglun’a Decoction(ZLD)on the apoptosisrelated factors of synovial cells in collagen-induced arthritis(CIA)model rats.Methods:The CIA rats model was established with bovine typeⅡcollagen.After SD rats were successfully modeled,CIA model rats were randomly divided into model group,ZLD high,medium,low dose group,and methotrexate(MTX)group.All rats were given intragastric administration for six weeks,the changes of rat body weight,right hind paw thickness and arthritis index(AI)were recorded before and after administration.The pathological observation on the ankle joint structure was observed with light microscope and the expression of Bax,Bcl-2 and caspase-3 were detected by immunohistochemical method in synovial tissues.Results:Compared with the CIA group,the ZLD reduced the paw thickness and AI in rats,and improved the pathological changes of ankle joints.Compared with the CIA group,the ZLD high,middle dose group increased the level of Bax and caspase-3(P<0.01),and reduced the protein expression level of Bcl-2(P<0.01)in synovial tissues.Conclusion:The ZLD may induce CIA rats synovial cells apoptosis through regulating expression of apoptosis-related factors,and alleviate joint injury in CIA rats.
