摘要
目的为了解牙龈卟啉单胞菌(Porphyromonas gingivalis,Pg)及程序性细胞死亡因子4(PDCD4)在食管鳞状细胞癌(ESCC)中的表达,探讨其表达的相关性及与ESCC患者预后的关系。方法采用免疫组织化学SP法检测Pg及PDCD4在30例食管鳞癌组织及其癌旁组织中的表达,采用卡方检验、Spearman相关分析、Kaplan-Meier方法对食管癌组织及癌旁正常组织中PDCD4、Pg的表达及其与临床病理特征、生存期的关系进行分析。结果Pg在食管癌组织中的阳性率为53.3%(16/30),显著高于癌旁组织的20.0%(6/30);PDCD4在食管癌组织中的阳性率为40.0%(12/30),显著低于癌旁组织的100%(30/30),差异均具有统计学意义(P<0.05)。PDCD4在Pg阳性食管鳞癌组织中的阳性率(0/16)显著低于在Pg阴性食管鳞癌组织中的阳性率(85.7%,12/14)。Kaplan-Meier生存分析显示PDCD4阴性、Pg阳性患者的3 a生存率较低、预后较差。结论Pg高表达、PDCD4低表达与食管鳞癌的发生、发展密切相关,且Pg的高表达和PDCD4的低表达之间具有显著相关性,两者均可作为早期诊断及评估预后的指标。
Objective To understand the expression of Porphyromonas gingivalis(Pg)and programmed cell death factor 4(PDCD4)in esophageal squamous cell cancer(ESCC),to explore its expression correlation and relationship with the prognosis of ESCC patients.Methods Immunohistochemistry SP method was used to detect the expression of Pg and PDCD4 in ESCC and its adjacent tissues in 30 cases.Chi-square test and Kaplan-Meier method were used to analyze the expressions of PDCD4 and Pg in esophageal cancer tissues and adjacent normal tissues and their relationship with clinicopathological features and survival.Results The positive rate of Pg in esophageal cancer tissues was 53.3%(16/30),significantly higher than that in paracancer tissues(20.0%,6/30).The positive rate of PDCD4 in esophageal cancer tissues was 40.0%(12/30),significantly lower than that in paracancer tissues(100%,30/30),and the differences were statistically significant(P<0.05).The positive rate of PDCD4 in Pg-positive esophageal squamous cell carcinoma tissues(0/16)was significantly lower than that in Pg-negative esophageal squamous cell carcinoma tissues(85.7%,12/14).Kaplan Meier survival analysis showed that patients with PDCD4 negative and PG positive had lower 3-year survival rate and poor prognosis.Conclusion High Pg expression and low PDCD4 expression are closely related to the occurrence and development of esophageal squamous cell carcinoma,and there is a significant correlation between high Pg expression and low PDCD4 expression,both of which can be used as indicators for early diagnosis and prognosis evaluation.
