波斯顿蕨绿色小体及孢子体诱导与增殖优化 被引量:1

Induction and multiplication optimization of green globular bodies and sporophytes of Bostoniensis

摘要 以2个品种波斯顿蕨试管苗根状茎茎尖为外植体,研究绿色小体和孢子体的诱导分化与增殖,开展孢子体叶状体芽和叶芽成苗及生根试验.结果表明:MS+6-BA0.50 mg/L为金叶波斯顿蕨绿色小体最佳诱导分化培养基,绿色小体(GGB)诱导率为87.60%;绿叶波斯顿蕨绿色小体最佳诱导分化培养基为MS+6-BA0.75 mg/L,GGB诱导率为88.24%;MS+6-BA1.00 mg/L为2种波斯顿蕨最佳绿色小体增殖培养基,金叶波斯顿蕨和绿叶波斯顿蕨绿色小体增殖倍数分别为7.30和7.94.MS+6-BA1.0 mg/L+NAA0.3 mg/L为金叶波斯顿蕨孢子体最佳诱导培养基,诱导率为92.0%;MS+6-BA1.2 mg/L+NAA0.4 mg/L为绿叶波斯顿蕨孢子体最佳诱导培养基,诱导率为90.6%;MS+KT0.5 mg/L+NAA0.2 mg/L为波斯顿蕨最佳芽成苗培养基,金叶波斯顿蕨和绿叶波斯顿蕨的叶芽成苗率分别为96.0%和92.4%.波斯顿蕨最佳的生根培养基为MS+IBA0.2 mg/L+2,4-D0.2 mg/L+0.1%Ac. The shoots and stems of two species of Boston fern in vitro were used as explants to study the differentiation and proliferation of green corpuscles and sporophytes.The spore body buds and leaf buds were seeded and rooted.The results show:MS+6-BA 0.5 mg/L was the best induction differentiation medium for the fern green body of Golden Leaf,and the GGB induction rate was 87.60%.The optimal differentiation medium for green leaf Boston fern green body was MS+6-BA 0.75 mg/L,and the GGB induction rate was 88.24%;MS+6-BA 1.00 mg/L is the best green body growth medium for two Boston ferns.The multiplication ratios of gold leaf Boston fern and green leaf Boston fern green bodies are 7.30 and 7.94,respectively.MS+6-BA 1.0 mg/L+NAA 0.3 mg/L was the best induction medium for the fern spores of Boston,and the induction rate was 92%.MS+6-BA 1.2 mg/L+NAA 0.4 mg/L It is the best induction medium for the fern spores of the green leaf,and the induction rate is 90.6%.MS+KT 0.5mg/L+NAA 0.2mg/L was the best bud seedling culture medium for Boston fern,and the leaf germination rate of golden leaf Boston fern and green leaf Boston fern was 96.0%and 92.4%,respectively.The best rooting medium for Boston fern is MS+IBA 0.2 mg/L+2,4-D 0.2 mg/L+0.1%Ac.
