摘要
应用近红外光谱测定及细菌培养平板计数的方法建立水分活度为0.1的青金桔原粉中大肠杆菌、酵母、霉菌总数的快速检测模型。通过考察温度0℃、27℃、35℃的不同保存条件下青金桔粉中霉菌、酵母、大肠杆菌的近红外光谱以及微生物菌数变化,建立青金桔粉中菌落数的近红外模型。结果表明:霉菌总数的校正和预测模型的有效决定系数分别为0.9811和0.9823,酵母菌总数的校正和预测模型有效决定系数分别为0.8794和0.8264,大肠杆菌总数的校正和预测模型的有效决定系数分别为0.9420和0.9904,实验研究的模型对青金桔粉中大肠杆菌和霉菌总数的检测准确性较高,可用于快速、无损检测。
The method of near infrared spectroscopy and bacterial culture plate count method was used to determine the total bacterial activity of kumquat powder.First,weighed the amount of green kumquat powder,in the water activity of 0.1 under the conditions of mixing and evenly spread in a dry culture dish,the temperature 0℃,27℃,35℃under different preservation conditions for parallel determination,the establishment of the total number of moulds,yeasts and Escherichia coli in the samples were determined by plate colony counting method,and the total number of colonies in the green kumquat was established by the near-infrared spectroscopy of the moulds,yeast,E.coli.The correlation coefficients of calibration and prediction for mould were 0.9811 and 0.9823 respectively.The correlation coefficients of calibration and prediction for yeast were 0.8794 and 0.8264 respectively.The correlation coefficients of calibration and prediction for E.coli were 0.9420 and 0.9904 respectively.Variable near infrared model for E.coli and mould can achieve the fast,non-destructive testing of bacteria in the green kumquat powder.
