摘要
羊毛是养羊业的主要产品之一,其是由皮肤内的附属器官毛囊发育而来,其中弯曲度是衡量其质量的重要指标之一。为了解NF-κB家族各亚基(NFκB1、NFκB2、RELA、RELB和REL)在不同品种绵羊背部皮肤中基因水平的表达差异,分析其在不同品种绵羊背部皮肤中与形成毛发性状差异相关的关键性基因,选取美利奴羊和晋中绵羊2个不同品种的绵羊作为研究对象,利用实时荧光定量PCR方法检测不同品种绵羊背部皮肤中NF-κB各亚基的表达水平。结果显示,NF-κB各亚基在美利奴羊和晋中绵羊背部皮肤中的相对表达量均有差异,除RELA外,其余各亚基mRNA的相对表达量美利奴羊均高于晋中绵羊;美利奴羊皮肤中,NFκB2 mRNA相对表达量最高,RELA mRNA次之;晋中绵羊皮肤中,RELA mRNA相对表达量最高,NFκB2 mRNA次之。结果揭示,NFκB2可能对于羊毛弯曲性状的形成具有促进作用,而RELA可能对于羊毛弯曲性状的形成具有抑制作用;NFκB2与RELA适当的相对表达水平可能对于毛发弯曲性状的形成起重要的调控作用。
Wool is one of the main products of sheep farming.It is developed from the hair follicles of the accessory organs in the skin.The bending is one of the important indicators to measure its quality.To understand the expressed differences at gene level of NF-κB family subunits(NFκB1,NFκB2,RELA,RELB and REL)in the dorsal skin of different breeds of sheep,the key gene related to the formation of different hair traits in the dorsal skin of different breeds of sheep were analyzed.Two different breeds of sheep,Merino sheep and Jinzhong sheep,were selected as research objects,the expression level of NF-κB subunits in the dorsal skin of different breeds of sheep were detected by real-time fluorescent quantitative PCR.The results showed that the relative expressions of NF-κB family subunits in the dorsal skin of Jinzhong sheep and Merino sheep were different.Except for RELA,the relative expression level of the mRNA of all subunits in Merino sheep were higher than those in Jinzhong sheep.Among them,the relative expression of NFκB2 mRNA in the skin of Merino sheep was the highest,followed by RELA mRNA.The relative expression of RELA mRNA in the skin of Jinzhong sheep was the highest,followed by NFκB2 mRNA.The results suggest that NFκB2 may promote the formation of wools'bending trait,while RELA may inhibit the formation of wools'bending trait.Linking the two results suggests that the appropriate relative expression level of NFκB2 and RELA may play an important regulatory role for the formation of hair bending trait.
