摘要
目的通过网络药理学方法,分析五味子治疗哮喘的有效活性成分及相关作用靶点.方法应用中药系统药理学数据库(TCMSP)整理五味子的活性成分及预测相应的作用靶点,采用STRING数据库与Cytoscape软件构建"药物-活性成分-靶点"网络和蛋白质相互作用网络,并通过相关生物途径进一步筛选.结果通过口服生物利用度(OB)和类药性(DL)两个参数筛选出8个有效活性成分和56个作用靶点.通过TTD数据库收集哮喘相关的基因共132个,筛选出五味子与哮喘相关的候选作用靶点共10个,其中最具相关性的化合物为醇乙、酯乙、戈米辛R.结论五味子可能是通过调控NF-κB、PPAR、IL-17等信号通路抑制促炎因子的基因表达,进而发挥治疗哮喘的作用,其主要靶标可能是Pparg、Ptgs2、Nos2,其主要活性成分为醇乙、酯乙、戈米辛R.
Objective To analyze the active components of Schisandra chinensis and the related targets for asthma treatment through network pharmacology method. Method The active components of Schisandra chinensis were collected and their targets were predicted by TCMSP.The "drug-active components-targets"network and proteinprotein interaction network were constructed by using STRING database and Cytoscape software,and the relevant biological pathways were also analyzed.Results 8 active components and 56 targets were obtained by OB and DL screening.132 genes related to asthma were collected from TTD database.There were 10 related candidate targets,and the most relevant component were Schisandrol B,Schizantherin B and Gomisin R. Conclusion Schisandra chinensis may inhibit the gene expression of pro-inflammatory factors by regulating NF-κB,PPAR,IL-17 and other signaling pathways,and play a role in the treatment of asthma.The main targets may be Pparg,Ptgs2 and Nos2,and the main active component are Schisandrol B,Schizantherin B and Gomisin R.
