摘要
目的通过网络药理学方法探讨参苓白术散对慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)和溃疡性结肠炎(ulcerative colitis,UC)"异病同治"的作用机制。方法在TCMSP和TCMID数据库检索参苓白术散中10味中药的化学成分和作用靶点,在OMIM、GeneCards、TTD、DrugBank数据库中查找COPD和UC的疾病靶点,运用Venny 2.1.0工具获取参苓白术散、COPD及UC三者的交集靶点,在STRING数据库中分析蛋白互作关系,并运用Cytoscape 3.7.2软件构建药物活性成分-靶点-疾病网络图以及核心靶点网络图。利用DAVID6.8数据库进行基因本体论(GO)功能分析和京都基因与基因组百科全书(KEGG)通路富集分析。结果根据筛选条件检索得到参苓白术散活性成分118个,作用靶点195个,COPD靶点1026个,UC靶点835个,交集靶点52个,GO功能分析得到生物过程115个、细胞组成13个、分子功能22个,KEGG富集分析获得95条信号通路,主要涉及HIF-1信号通路、MAPK信号通路、NF-κB信号通路、TNF信号通路、TCR信号通路、PI3K-Akt信号通路。结论参苓白术散对COPD和UC"异病同治"的主要机制涉及免疫炎症、代谢以及氧化应激相关信号通路,为参苓白术散在临床中治疗COPD和UC提供理论依据。
Objective To investigate the mechanism of Shenling Baizhu Powder(参苓白术散)in treating chronic obstructive pulmonary disease(COPD)and ulcerative colitis(UC)by the network pharmacology.Methods The chemical constituents and action targets of Shenling Baizhu Powder were searched on the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)and Traditional Chinese Medicine Integrated Database(TCMID)databases.The disease-related targets of COPD and UC were found in OMIM,Genecards,TTD and Drugbank databases and we use Venny 2.1.0 tool obtained the intersection targets of Shenling Baizhu Powder,COPD and UC was constructed by using Cytoscape 3.7.2.The protein interaction was analyzed in STRING database,and the network diagram of active components-target-disease and the core target network was constructed by using Cytoscape 3.7.2 software.DAVID 6.8 database was used to analyze GO function and KEGG pathway enrichment.Results 118 active components,195 action targets,1026 COPD targets,835 UC targets were found according to the screening conditions,and 52 intersection targets were obtained.115 biological processes,13 cell composition and 22 molecular functions were obtained by GO function analysis.95 signal pathways were obtained by KEGG enrichment analysis,which mainly involve HIF-1,MAPK,NF-κB,TNF,TCR and PI3 K-Akt signaling pathway.Conclusion The main mechanism of Shenling Baizhu Powder in treating COPD and UC is related to immune inflammation,metabolism and oxidative stress related signaling pathways,which provides a theoretical basis for the clinical treatment of COPD and UC.
