摘要
目的探索高频中药治疗代谢综合征(Metabolic Syndrome,MS)的核心作用靶点、主要活性成分及作用机制。方法以黄芪、茯苓、丹参、泽泻、山楂、半夏、大黄、白术、黄连、川芎为关键词分别检索TCMSP数据库,按照OB≥30%,DL≥0.18的条件筛选,获得高频中药的活性成分及其作用靶点;通过DrugBank、Pharmgkb、CTD、TTD数据库检索MS的疾病靶标;将预测到的成分及MS的疾病靶标运用Cytoscape软件进行MS靶点-中药成分的网络构建,筛选核心靶点及主要活性成分,DAVID数据库行KEGG通路富集。结果①高频中药成分可能作用于PPARG、ESR、eNOS、ADRB2、PTGS1、PTGS2、AR等7个核心靶点,通过调节脂质代谢、调节血压水平、减轻胰岛素抵抗等达到治疗MS的效果。②高频中药治疗MS的主要活性成分为黄酮类及醌类化合物。③高频中药可能通过调控上游通路胰岛素抵抗通路,下游通路涉及PPAR、雌激素、TNF等信号通路及花生四烯酸代谢、脂肪细胞的脂解等作用达到治疗MS的作用。结论高频中药治疗代谢综合征呈现出多成分、多靶点、多通路的作用特点,其主要活性成分可通过调节脂质代谢、调节血压水平、减轻胰岛素抵抗等参与对代谢综合征不同危险因素的调控,进而影响代谢综合征的发展。
Objective To explore the core target, main active components and mechanism of high frequency traditional Chinese medicine in the treatment of metabolic syndrome. Methods TCMSP databases were retrieved using Radix Astragali seu Hedysari, Poria, Radix Salviae Miltiorrhizae, Rhizoma Alismatis, Fructus Crataegi, Rhizoma Pinelliae,Radix et Rhizoma Rhei, Rhizoma Atractylodis Macrocephalae, Rhizoma Coptidis, Rhizoma Ligustici Chuanxiong as keywords. According to the condition that OB is greater than 30% and DL is greater than 0.18, the chemical componentsand action targets of high-frequency traditional Chinese medicine were obtained. Then the disease target of metabolicsyndrome was retrieved by DrugBank Pharmgkb, CTD, and TTD database. The predicted components and the diseasetargets of metabolic syndrome were constructed into component-target network by using Cytoscape software to screen thecore target and its main active components and the relevant pathways were set up by using BinGO and DAVID. Thepotential active components and their mechanisms are explored by means of network and pathway. Results(1) Thecomponents of high-frequency traditional Chinese medicine may act on 7 core target proteins, such as PPARG, ESR,eNOS, ADRB2, PTGS1, PTGS2, AR, etc. to regulate lipid metabolism, regulate blood pressure level, reduce insulinresistance toachieve the effect of the treatment of metabolic syndrome.(2)The main active components of TCM treatmentof metabolic syndrome are flavonoids and quinines.(3)The main pathway of TCM treatment of metabolic syndrome isPPAR signaling pathway, Estrogen signaling pathway, TNF signaling pathway, arachidonic acid metabolism, regulationof lipolysis in adipocytes, insulin resistance. Conclusion High frequency traditional Chinese medicine for metabolicsyndrome is characterized by multiple components, multiple targets and multiple pathways. Its main active componentscan participate in the regulation of different risk factors of metabolic syndrome by regulating lipid metabolism, bloodpressure level, weight loss and insulin resistance. Thus, it can affect the development of metabolic syndrome.
