摘要
从定量对比角度分别利用浊度法、吸光度法、细胞计数法、紫外分光光度法对小球藻的破碎率进行了检测。先培养小球藻至实验所需数目浓度,再采用超声波法破碎小球藻。四种检测方法的检测结果表明:超声破碎后产生大量的小球藻碎片严重干扰了浊度法的检测结果;吸光度法的测量优于浊度法,利用吸光度法在680 nm波长下检测小球藻破碎率,其结果具有一定的规律性与参考价值;紫外分光光度法的测量结果较为理想,且操作简单;细胞计数法虽然操作复杂,但是能可靠地检测出破碎率。当超声功率为360 W、破碎时间为5 min时,小球藻破碎率为3%;当超声功率为1080 W,破碎时间为60 min时,小球藻破碎率则达到83.9%。
Four quantification indicators including the turbidity,visible light absorbance,cell counting and UV absorbance for cell disrupture of Chlorella vulgaris by ultrasonic were compared.Chlorella vulgaris was firstly cultivated and then ruptured by ultrasonic.The disrupture proportion was detected by the four indicators respectively.The results of turbidity were seriously interfered due to the large amount of cell debris produced after ultrasonic treatment.The visible light absorbance was more suitable than the turbidity.Absorbance at 680 nm had a reference for detecting the cell disrupture of Chlorella vulgaris.UV absorbance was much better for its convenience and detection efficiency.Cell counting was the most reliable indicator due to its capability of detecting specific disrupture proportion.According to the cell counting,the cell disrupture of Chlorella vulgaris was 3%with the ultrasonic power of 360 W and the treatment time of 5 min.It was 83.9%with the ultrasonic power of 1080 W and the treatment time of 60 min.
