CT定位大鼠单侧放射性肺损伤模型的构建

Construction of Animal Models of Unilateral Radiation-induced Lung Injury in Rats by CT-simulation

摘要 目的建立放射性肺损伤大鼠模型。方法雄性SD大鼠35只,随机分为对照组5只和模型组30只,模型组CT定位确定照射野后右胸一次性照射20 Gy,照射后第10、60、70、80 d处死大鼠后取肺组织,采用HE染色、Masson染色观察肺组织肺炎症分级及纤维化程度。结果模型组大鼠有6只在照射后死亡,照射后大鼠肺组织表现出不同程度的炎性细胞浸润及纤维化。肺组织炎症分级:对照组5只均为0级,模型组1级11只、2级12只、3级1只;肺纤维化分级:对照组5只均为0级,模型组0级2只、1级5只、2级10只、3级7只。结论通过CT模拟定位,20 Gy一次性照射大鼠右胸可成功构建放射性肺损伤动物模型,为研究放射性肺损伤发生机制提供一定基础。 Objective To establish rats model of radiation-induced lung injury.Methods 35 male SD rats were randomly divided into two groups:blank control group(n=5)and radiation group(n=30).After anesthesia,the irradiation field was determined by CT localizaion-simulation and the right chests of rats were irradiated with 20 Gy at one time.The 35rats were sacrificed at different time points and the lung tissues were taken out from rats.The severity of pulmonary inflammation and fibrosis were observed under the microscope by the methods of HE and Masson staining.Results After irradiation,different degrees of inflammatory cell infiltration and fibrosis occurred in the lung tissue.The lung inflammation was graded as follows.5 rats of the control group were grade 0,1 rat in the model group was grade1,12 rats were grade 2,1 rat was grade3.Pulmonary fibrosis was graded as follows,5 rats in the control group were all grade 0.In the model group,2 rats was grade 0,5 rats was grade 1,10 rats was grade 2,and 7 rats was grade 3.Conclusion By means of CT localization-simulation,rat models of radiation-induced lung injury can be successfully constructed by 20 Gy irradiation on the right chest,which provides a basis for studying the mechanism of radiation-induced lung injury.
