摘要
目的探讨转化生长因子-β(TGF-β)在放射性肺损伤大鼠肺组织中的表达及意义。方法选择无特定病原体(SPF)级健康Wistar大鼠35只,随机分为对照组(5只)、照射组(30只,1d,1、2、4、8、12周各5只)。在照射1d,1、2、4、8、12周获取标本,采用免疫组化法观察放射性肺损伤大鼠肺组织细胞TGF-β水平变化。结果照射组TGF-β在肺成纤维细胞中的表达高于对照组,差异有统计学意义(P=0.000),其损伤程度随时间的推移而增加。结论 TGF-β是肺成纤维细胞(FB)活化的最重要生物介质之一,阻断TGF-β通路及FB的活化,可能为减少照射后肺间质炎症和纤维化的程度带来新的希望。
Objective To investigate the expression of TGB-β on lung tissue of rats with radiation induced lung injury.Methods Thirty-five male Wistar rats were randomly divided into 7 groups(n=5):control group and treatment group(received irradiation on the the right hemithorax or sham right lung irradiation with single 30Gy,1day,1,2,4,8,and 12 weeks).The rats were sacrificed at 1 day,1,2,4,8,and 12 weeks after the irradiation,and lung tissues were separated.The expression of TGF-β was detected by immunohistochemistry.Results The expression of TGF-β in fibroblast(FB) significantly increased after irradiation,compared with control group(P=0.000).The degree of injury increased with the time passing.Conclusion TGF-β was an important biological mediator during the activation of fibroblast.We suspected that FB might be the target cell for prevention and cure the radiation induced lung injury.
