MDC1与YAP1在卵巢癌中的表达及与临床特征和预后的关系 被引量:3

Expression of MDC1 and YAP1 in patients with ovarian cancer and the correlation with clinical characteristics and prognosis

摘要 目的探讨卵巢癌患者卵巢癌组织中Yes相关转录调节因子1(YAP1)、DNA损伤介质检查点1(MDC1)的表达情况,并分析其与患者临床特征和预后的关系。方法选取118例卵巢癌组织标本为卵巢癌组,56例卵巢正常组织标本为对照组,采用免疫组化法检测YAP1、MDC1在卵巢癌组织及正常卵巢组织中的表达情况,并分析其与临床特征及预后的关系。结果YAP1、MDC1在卵巢癌组织中的阳性表达率均明显高于正常卵巢组织,差异均有统计学意义(P﹤0.01)。分化程度为G1级、FIGO分期为Ⅲ+Ⅳ期、有淋巴结转移的卵巢癌患者的卵巢癌组织中YAP1、MDC1阳性表达率均分别明显高于分化程度为G2~3级、FIGO分期为Ⅰ+Ⅱ期、无淋巴结转移的卵巢癌患者,差异均有统计学意义(P﹤0.01)。死亡组患者的卵巢癌组织中YAP1、MDC1阳性表达率均明显高于生存组,差异均有统计学意义(P﹤0.01)。YAP1、MDC1在卵巢癌组织中表达无相关性(r=0.126,P=0.213)。结论卵巢癌患者卵巢组织中YAP1、MDC1均呈高表达,且与患者临床分期、分化程度、淋巴结转移及预后有关,可能参与了卵巢癌的发生与发展,可以用于卵巢癌的辅助诊断。 Objective To investigate the expression of Yes1 associated transcriptional regulator(YAP1)and mediator of DNA damage checkpoint 1(MDC1)in ovarian tissue of patients with ovarian cancer and the correlation with clinical characteristics and prognosis.Method A total of 118 specimens of ovarian cancer and 56 cases of normal tissues were collected from corresponding patients as ovarian cancer group and control group,respectively.YAP1 and MDC1 expression in these specimens were detected by immunohistochemistry,besides,the relationship between their expression and clinical characteristics and the prognosis were analyzed.Result The positive expression level of YAP1 and MDC1 in ovarian cancer tissues were significantly higher than those in normal ovarian tissues(P<0.01),and were also higher in ovarian cancer tissues with degree of differentiation at G1,FIGO stage III+IV,and positive lymph node metastasis compared to those with differentiation of G2-3,FIGO stage I+II,and negative lymph node metastasis,indicating statistically significant differences(P<0.01).The positive expression rates of YAP1 and MDC1 were significantly higher in dead cases than those in survivors,showing statistical difference(P<0.01).Nonetheless,no correlation between the positive expression level of YAP1 and MDC1 in ovarian cancer tissues was observed(r=0.126,P=0.213).Conclusion The expression of YAP1 and MDC1 in ovarian tissue of patients with ovarian cancer are high,and are related to clinical stage,degree of differentiation,lymph node metastasis and prognosis.Their expression may be involved in the occurrence and development of ovarian cancer and thus is useful in the adjuvant diagnosis of ovarian cancer.
