ANXA2、STAT3、VEGF在乳腺癌中的表达及与肿瘤转移的关系 被引量:15

The expression of ANXA2,STAT3 and VEGF in breast cancer and their relationship with breast cancer metastasis

摘要 目的探讨膜联蛋白A2(ANXA2)、信号转导与转录激活因子3(STAT3)、血管内皮生长因子(VEGF)在乳腺癌组织中的表达情况及与肿瘤转移的关系。方法选取98例乳腺癌患者,收集其术中切除的乳腺癌组织及对应的癌旁正常组织,采用实时定量逆转录聚合酶链反应(RT-PCR)检测乳腺癌组织及癌旁正常组织中ANXA2、STAT3、VEGF mRNA的相对表达量,以及ANXA2、STAT3、VEGF的相关性,比较不同临床特征乳腺癌患者乳腺癌组织中ANXA2、STAT3、VEGF mRNA的相对表达量。结果乳腺癌组织中ANXA2、STAT3、VEGF mRNA的相对表达量均明显高于癌旁正常组织,差异均有统计学意义(P﹤0.05)。乳腺癌组织中ANXA2与STAT3、VEGF的表达均呈正相关(r=0.571、0.522,P﹤0.05),STAT3与VEGF的表达呈正相关(r=0.604,P﹤0.05)。不同TNM分期、淋巴结转移情况、远处转移情况、人表皮生长因子受体2(HER2)表达情况、上皮钙黏素(E-cadherin)表达情况、神经钙黏素(N-cadherin)表达情况、基质金属蛋白酶2(MMP2)表达情况和MMP9表达情况乳腺癌患者乳腺癌组织中ANXA2、STAT3、VEGF mRNA的相对表达量比较,差异均有统计学意义(P﹤0.05)。结论ANXA2、STAT3、VEGF在乳腺癌组织中表达上调,且与乳腺癌的转移密切相关。 Objective To investigate the expression of annexin A2(ANXA2),signal transduction and activator of transcription 3(STAT3)and vascular endothelial growth factor(VEGF)in breast cancer and their relationship with breast cancer metastasis.Method The research included 98 patients with breast cancer whose breast cancer tissues excised during operation and corresponding normal breast tissues adjacent to cancer lesions were collected as test samples.The expressions of ANXA2,STAT3 and VEGF mRNA in these tissue samples were detected by real-time quantitative reverse transcription polymerase chain reaction(RT-PCR).The correlation among the three markers and the association of each with the pathological characteristics and metastasis related indicators of breast cancer patients were analyzed.Result The relative expression levels of ANXA2,STAT3 and VEGF mRNA in breast cancer tissues were significantly higher than those in adjacent normal tissues(P<0.05).The expression of ANXA2 was positively correlated with STAT3 and VEGF in breast cancer,respectively(r=0.571,0.522,P<0.05),besides,STAT3 expression was also positively related to VEGF expression(r=0.604,P<0.05).There were significant differences observed among the relative expression of ANXA2,STAT3 and VEGF mRNA in breast cancer tissues with respect to TNM staging,lymph node metastasis,distant metastasis,human epidermal growth factor receptor 2(HER2)expression,E-cadherin expression,N-cadherin expression,matrix metalloproteinase 2(MMP2)expression and MMP9 expression(P<0.05).Conclusion The expression of ANXA2,STAT3 and VEGF mRNA are up-regulated in breast cancer tissues,which are positively correlated with the metastasis of breast cancer.
