三氯生对斑马鱼卵巢抗氧化和凋亡相关基因表达的影响 被引量:2

摘要 为了探讨三氯生(TCS)对硬骨鱼类性腺损伤作用的分子机制,通过半静态水体暴露法对斑马鱼(Danio rerio)进行染毒,将斑马鱼暴露在2种不同TCS浓度(0.034 mg/L和0.068 mg/L)溶液中,同时设置空白对照组,42 d后,利用实时荧光定量PCR(RT-qPCR)技术,探究TCS对斑马鱼卵巢抗氧化及凋亡相关基因表达的影响。结果发现:与空白对照相比,SOD、CAT和GPx1a基因在0.068 mg/L TCS处理组表达水平显著下调,GPx1和MT-2基因在0.034 mg/L浓度组表达显著上调;Bcl-2基因在0.068 mg/L浓度组表达显著下调,p53基因在0.034 mg/L浓度组表达极显著上调,Bax在TCS处理组中均显著上调。以上研究结果表明:较高浓度TCS显著影响了斑马鱼卵巢抗氧化和凋亡相关基因的表达,从而产生了氧化损伤和加速细胞凋亡的发生。 Danio rerio were exposed to the blank control and two different concentrations(0.034 mg/L and 0.068 mg/L)of TCS for a 42 d semi-static exposure to investigate the molecular mechanism of triclosan(TCS)on gonadotrophic damage in teleost fish.Then mRNA levels of antioxidation and apoptosis-related genes in zebrafish ovary were determined using real time quantitative PCR(PT-qPCR)technique.The results showed that mRNA expression levels of SOD,CAT,and GPx1 in 0.068 mg/L TCS treated group were significantly decreased compared with the control,while GPx1 and MT-2 mRNA levels were significantly increased in 0.034mg/L TCS treated group.Bcl-2 mRNA level in 0.068 mg/L TCS treated group was significantly decreased,while p53 gene in 0.034 mg/L TCS treated group was significantly increased and Bax gene in treated groups were significantly increased.These results showed that higher concentration of TCS significantly affected the mRNA expression of antioxidant and apoptotic-related genes in zebrafish ovary,which resulted in oxidative damage and accelerating cell apoptosis.
