摘要
目的:观察沙榆油对急性放射性皮肤损伤模型皮肤损伤的防治效果及对凋亡相关基因bcl-2和bax表达的影响。方法:雌性SD大鼠40只,均行臀部照射野皮肤脱毛,随机选取10只为空白组,不做特殊处理,其余30只进行急性放射性皮肤损伤造模后随机分为模型组、治疗组和对照组,每组10只。从造模次日起均以生理盐水清洁放射野皮肤,治疗组清洁后外施沙榆油,对照组清洁后外施湿润烧伤膏,模型组不做处理,每天1次。比较各组大鼠照射野皮肤变化,免疫组织化学检测bcl-2和bax的表达情况。结果:造模3组大鼠均发生急性放射性皮肤损伤,治疗组较轻,模型组最重,对照组较重。第14天,与空白组比较,其余3组大鼠皮损组织中bcl-2蛋白均呈低表达或阴性,差异均有统计学意义(P<0.05)。第30天,与空白组比较,治疗组bcl-2蛋白表达明显升高,差异无统计学意义(P>0.05);治疗组bcl-2蛋白表达显著高于模型组、对照组(P<0.05);模型组、对照组bcl-2蛋白表达上升缓慢,均显著低于空白组(P<0.05)。第14天,与空白组比较,其余3组bax蛋白均呈阳性(++)至强阳性,差异均有统计学意义(P<0.05);治疗组与模型组比较,差异有统计学意义(P<0.05)。第30天,治疗组bax蛋白表达进一步减弱,与模型组、对照组比较,差异均有统计学意义(P<0.05);模型组、对照组bax蛋白的表达仍然较强,与空白组比较,差异有统计学意义(P<0.05)。结论:沙榆油可以减轻放射性皮肤损伤的程度,促进放射性皮肤损伤的恢复;其机制可能与其增强皮损细胞中bcl-2蛋白的表达、降低bax基因的表达、抑制细胞凋亡的作用有关。
Objective:To observe the effects of Shayu Paste on the skin damage of acute radiation-induced skin injury model and the effects of apoptosis-related genes bcl-2 and bax.Methods:Forty female Sprague-Dawley rats were depilated from the skin of the buttocks.Ten randomly selected groups were randomly selected and no special treatment was performed.The other 30 were randomly divided into model group,treatment group and the control group was 10 in each group.The skin of the radiation field was cleaned with normal saline from the next day after the model was established.The treatment group was treated with Shayu Paste after cleansing.The control group was cleaned and applied with moist burn cream.The model group was not treated once a day.The skin changes of the rats in each group were compared and the expression of bcl-2 and bax were detected by immunohistochemistry.RESULTS:Acute radiation-induced skin lesions occurred in the 3 groups of rats.The treatment group was lighter,the model group was the heaviest,and the control group was heavier.On the 14 th day,compared with the blank group,the bcl-2 protein in the skin lesions of the other three groups was low or negative,and the difference was statistically significant(P<0.05).On the 30 th day,compared with the blank group,the expression of bcl-2 protein in the treatment group was significantly increased,and the difference was not statistically significant(P>0.05).The expression of bcl-2 protein in the treatment group was significantly higher than that in the model group and the control group(P<0.05).The expression of bcl-2 protein in the model group and the control group increased slowly,which were significantly lower than that in the blank group(P<0.05).On the 14 th day,compared with the blank group,the other three groups of bax protein were positive(++) strong positive,the difference was statistically significant(P<0.05);the treatment group compared with the model group,the difference was statistically significant(P<0.05).On the 30 th day,the expression of bax protein in the treatment group was further attenuated.Compared with the model group and the control group,the difference was statistically significant(P<0.05).The expression of bax protein in the model group and the control group remained strong,compared with the blank group.The difference was statistically significant(P<0.05).Conclusion:Shayu Paste can reduce the degree of radiation-induced skin damage and promote the recovery of radiation-induced skin damage.The mechanism may be related to the enhancement of bcl-2 protein expression,the decrease of bax gene expression and the inhibition of apoptosis.
