摘要
目的研究大鼠急性β射线皮肤损伤创面愈合过程中酪氨酸激酶免疫球蛋白和表皮生长因子同源受体-2(Tie-2)mRNA的表达情况。方法将54只雄性SD清洁级大鼠随机分为3组。照射组(=24),直线加速器产生射线(45Gy)单次照射大鼠臀部皮肤40 mm×40 mm,建立急性射线皮肤损伤模型;烧伤组(=24),将直径30mm铜柱于100℃沸水中放置15 min,取出后在无外界压力下置于大鼠臀部皮肤8 s,建立深Ⅱ°热力烧伤模型;对照组(=6),正常大鼠。取不同时期创面皮肤组织,采用原位逆转录聚合酶链反应(RT-PCR),检测不同时点各组大鼠创面组织中Tie-2mRNA的表达。结果 A组大鼠照射后第2天皮肤出现充血性红斑,2周后脱毛,4周后出现溃疡创面,5周后创面仍未愈合。B组大鼠烧伤后创面即刻出现水泡,3d后肉芽增生,3~4周后创面瘢痕愈合。大鼠正常皮肤内Tie-2mRNA为可疑阳性;A组大鼠创面Tie-2mRNA表达伤后1周略有升高,并持续低表达;B组Tie-2 mRNA明显上调,2周达峰,3周时仍高于A组(均<0.05)。结论急性射线皮肤损伤创面愈合过程中Tie-2的持续低表达,不能形成峰值,可能是急性射线皮肤损伤创面难愈合的机制之一。
Objective To investigate the dynamic expression of tyrosine kinase immunoglobulin and Tie-2 mRNA in coalescence among the rats with β-ray radiation skin injury.Methods Fifty-four male SD rats were divided into three groups.The model of acute radiation injury of skin in rats was replicated by 45 Gy β-ray to the skin radiation(A,n = 24);the copper column with 30 mm diameter was be placed in boiling water for 15 minutes,then it was placed on the skin of the hip eight seconds in the absence of outside pressure,to establish the model for deep second-degree burns(B,n=24);Six normal rats were served as normal control group(N,n=6).From different periods skin wounds,the expression of Tie-2 mRNA protein were respectively semi-quantitative checked by means of immunohistochemical technique SP.ResultsAmong group A,congestive erythema appeared two days after radiation;hair removal appeared two weeks after radiation;ulcers appeared four weeks after radiation;ulcers were not healed five weeks after radiation.Among group B,water vacuole turns up immediately;granulation proliferation three days later;scars were healed three of four weeks.Lower expression of Tie-2 mRNA was identified in the rat skin in normal control group.The Tie-2 mRNA value of the R group was 0.298±021,0.334±031,0.351±032,0.396±018 and 0.411±014;and the B group was 0.381±034,0.528±032,0.451±031,0.348±033 and 0.312±011.There were also statistical significances between the two groups(P0.05).ConclusionsThe continuing low expression of Tie-2 mRNA may be partially involved in the mechanism of retarded healing of acute β-ray radiation injury in skin wounds.
