膳食联合STZ诱导胰岛素抵抗的2型糖尿病大鼠模型的建立 被引量:18

摘要 目的建立具有外周胰岛素抵抗的早期2型糖尿病肥胖大鼠模型。方法4周龄SD雄性大鼠,高糖高脂诱导4周,剔除膳食肥胖抵抗性大鼠。8周后,膳食肥胖性大鼠分别以30、60 mg/kg剂量空腹给予链脲佐菌素(STZ)腹腔注射,以体质量、空腹血糖、空腹胰岛素及糖耐量作为监测指标进行分析。结果幼龄大鼠高糖高脂饲喂4周后,63%的受试大鼠出现膳食诱导性肥胖,8周后,出现外周胰岛素抵抗。STZ诱导后,30 mg/kg剂量组出现2型糖尿病的早期症状,60 mg/kg剂量组出现了I型糖尿病的症状。结论高糖高脂诱发大鼠胰岛素抵抗联合30 mg/kg STZ诱导胰岛细胞轻度损伤可以构建具有早期2型糖尿病症状的大鼠模型。 Objective To establish a early type 2 diabetic obese rat model with peripheral insulin resistance.Methods Four-week-old SD male rats were induced with high glucose and high fat for four weeks,and dietary obesity-resistant rats were excluded.Eight weeks later,dietary obesity rats were given intraperitoneal injection of streptozotocin at the doses of 30 mg/kg and 60 mg/kg,respectively.Body weight,fasting blood glucose,fasting insulin and glucose tolerance were taken as monitoring indicators for analysis.Results After 4 weeks of high glucose and high fat induction,63%of the young rats developed dietary-induced obesity,and peripheral insulin resistance was observed after 8 weeks.After the injection of streptozotocin,the early symptoms of type 2 diabetes were found in the low-dose group and the symptoms of type 1 diabetes were found in the high-dose group.Conclusion The rat model of early type 2 diabetes with insulin resistance can be established by high glucose and high fat combined with low dose STZ at 30 mg/kg.
