摘要
从河西走廊敦煌地区盐碱土壤中筛选出一株对多种植物病原真菌均具有良好拮抗效果的放线菌DA4-3-12,对其最优发酵条件及稳定性进行探究,并对活性物质进行初步分离.采用平板涂布法确定最优培养基,琼脂扩散法筛选拮抗植病真菌菌株,单因素试验和正交试验确定其最优培养条件及稳定性,有机溶剂萃取法和大孔吸附树脂吸附法对活性物质进行初步分离并提高其抑菌活性.结果显示,菌株DA4-3-12为植病真菌拮抗放线菌,最优发酵条件为:发酵时间156 h、接种量8%、装液量90/250 mL、起始pH 10.菌株在不同酸碱、紫外照射时间、温度、贮存时间条件下均表现出良好的稳定性.抑菌活性物质为脂溶性物质,有机溶剂氯仿及大孔吸附树脂HP-20均可对其进行初步分离.以上结果说明菌株DA4-3-12对植物病原真菌有良好的抑制效果,有较好的实际应用前景.
A strain of actinomycetes DA4-3-12 with good antagonistic effect against various plant pathogenic fungi was selected from the salt-alkali soil in Dunhuang area of the Hexi corridor.The optimal fermentation condition and product stability were investigated,and therefore the active substance of strain DA4-3-12 was preliminarily separated.Plate coating method was used for the determination of optimal medium,AGAR diffusion method was used to screen the strains of antagonistic phytophytic fungi,single factor test and orthogonal test were used for the determination of the optimal culture condition and product,and organic solvent extraction and macroporous adsorption resin were used for separation the active substance and improvement the antibacterial activity.The results showed that actinomycete strain DA4-3-12 is antagonistic plant fungus actinomycete and the optimal fermentation conditions are:fermentation time 156 h,inoculation quantity 8%,loading volume 90/250 mL,initial pH 10.The strain DA4-3-12 shows good stability under different pH,ultraviolet irradiation,temperature and storage time.The antibacterial active substance is lipid soluble substances,which can be separated by organic solvent chloroform and macroporous adsorption resin.Strain DA4-3-12 has a good inhibitory effect on plant pathogenic fungi and has a great prospect for practical application.
