摘要
该研究以皱纹盘鲍肠道中筛选得到的产琼胶酶弧菌(Vibrio)Y-AG3为研究对象,综合运用单因素试验、Plackett-Burman试验、最陡爬坡试验及Box-Behnken试验对其发酵产琼胶酶培养基组成进行优化。结果表明,最优培养基组成为NaCl 2%,CaCl20.1%,琼脂0.59%,蛋白胨0.2%,MgSO40.4%,酵母膏0.6%。在此最优培养基条件下,琼胶酶活力最高,为51.60 U/mL,比优化前提高了16.74%。
Using agarase-producing Vibrio Y-AG3 screened from the intestinal tract of Haliotis discus hannai as research object,the medium components of the agarase producing strain were optimized by single factor tests,Plackett-Burman tests,steepest climb test and Box-Behnken tests The results showed that the optimal medium components were NaCl 2%,CaCl20.1%,agar 0.59%,peptone 0.2%,MgSO40.4%and yeast extract 0.6%.Under the optimal medium,the activity of agarase was 51.60 U/ml,which was 16.74%higher than that before optimization.
