摘要
目的:研究青钱柳双瓜袋泡茶(PDC)对2型糖尿病小鼠血糖、血脂与肝肾功能的影响及机制。方法:采用高糖高脂饲料喂养4周后一次性腹腔注射链脲佐菌素(STZ,60 mg/kg)建立2型糖尿病小鼠模型,将造模成功的小鼠随机分为模型组、二甲双胍组(0.387 g/kg)和PDC高、中、低剂量组(9.1 g/kg、4.55 g/kg、2.275 g/kg),另设正常对照组,每组10只。分别灌胃给药4周后(正常对照组和模型组给予等量蒸馏水)测定各组空腹血糖(FBG)、血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、尿素氮(BUN)、肌酐(CREA)、低密度脂蛋白胆固醇(LDLC)、高密度脂蛋白胆固醇(HDLC)、总胆固醇(TC)、甘油三酯(TG)、过氧化氢酶(CAT)水平和肝脏组织中超氧化物歧化酶(SOD)、丙二醛(MDA)含量,采用苏木精—伊红(HE)染色法观察胰腺组织病理学形态。结果:与模型组比较,二甲双胍组和PDC中、高剂量组小鼠FBG水平显著降低,且PDC高剂量组FBG下降更明显(P<0.05),PDC低剂量组FBG水平无明显差异(P>0.05)。与模型组比较,二甲双胍组与PDC各剂量组血清和肝脏组织中ALT、AST、BUN、CREA、LDLC、TC、TG、MDA水平均明显降低(P<0.05或P<0.01),CAT、SOD、HDLC水平明显升高(P<0.05或P<0.01)。胰腺HE结果显示,模型组小鼠胰腺组织紊乱,胰岛数目较少,胰岛细胞出现变性、坏死现象;与模型组比较,PDC各剂量组小鼠胰腺形态恢复规则,胰岛细胞数目增加,排列整齐,细胞形态较为完整,其中PDC高剂量组胰腺组织病理变化改善最为明显。结论:PDC能明显降低2型糖尿病小鼠血糖、血脂,减轻胰岛细胞损伤,改善肝肾功能,可能与提高肝脏组织SOD活性、清除自由基、增强抗氧化能力有关。
Objective:To investigate the effects and mechanisms of Paliurus Duplex Cucumis tea bags(PDC)on the serum lipid,blood glucose,and liver and kidney function in type 2 diabetic mice.Methods:The type 2 diabetic mice model was established by feeding with high fat diet(HFD)for 4 weeks and intraperitoneal injection of streptozotocin(STZ)(60 mg/kg),then the mice were randomly divided into model group,metformin group(0.387 g/kg),and PDC high dose,medium dose and low dose groups(9.1 g/kg,4.55 g/kg,2.275 g/kg),with 10 mice in each group.Ten healthy mice were selected as normal control group.The fasting blood glucose(FBG),the levels of alanine transaminase(ALT),aspartate transaminase(AST),blood urea nitrogen(BUN),creatinine(CREA),low density lipoprotein cholesterol(LDLC),high-density lipoprotein cholesterol(HDLC),total cholesterol(TC),triglyceride(TG),catalase(CAT)in serum and the contents of superoxide dismutase(SOD)and methane dicarboxylic aldehyde(MDA)in liver tissues were detected.The pathological morphology of pancreatic was observed by hematoxylin-eosin(HE)staining.Results:Compared with the model group,the FBG level was significantly decreased in the metformin group and the PDC medium-and high-dose groups,and the reduce was more obvious in the PDC high dose group(P<0.05).There was no significant difference in FBG level between the PDC low dose group and the model group(P>0.05).Compared with the model group,the levels of ALT,AST,BUN,CREA,LDLC,TC,TG,and MDA in serum and liver tissues of the metformin group and PDC treatment groups were significantly reduced,while the CAT,SOD and HDLC levels were significantly increased(P<0.05 or P<0.01).HE results showed that the pancreatic tissue was disordered,the number of islets was less,and the islet cells appeared degeneration and necrosis in the model group,but the islet morphology recovered regular forms,the islet cells were increased,the cells were in ordered arrangement,and the cell morphology was relatively complete in PDC treatment groups.Among them,the pathological changes in the pancreas were the most significant in the PDC high dose group.Conclusion:PDC can significantly decrease blood glucose and serum lipid,improve antioxidant injury capability,ameliorate the hepatorenal function,protect islet cell from damage,and maintain the normal structure and function of the islet.
