摘要
以黑果枸杞实生苗茎段、下胚轴、叶片为外植体,设定不同浓度6-BA与NAA的组合,建立黑果枸杞外植体快繁体系。结果表明:黑果枸杞分化所需的外源激素浓度较低,以下胚轴、茎段及叶片为外植体分化不定芽的最佳激素浓度分别为6-BA 0.050 mg·L-1+NAA 0.020 mg·L-1、6-BA 0.100 mg·L-1+NAA 0.005 mg·L-1及6-BA 0.200 mg·L-1+NAA 0.005 mg·L-1,增殖数分别为3株·段-1、4株·段-1和5株·片-1。不定芽在MS培养基生根率达100%,长势良好。一年生黑果枸杞植株上、中及下部茎段和叶片均可按上述激素组合诱导长势健康的不定芽,上部的茎段及叶片诱导不定芽数明显高于下部。
Hypocotyls,stem segments and leaves of Lycium ruthenicum seedling were used as explant to set up a rapid propagation system with different concentrations of 6-BA and NAA.The results showed the exogenous hormones required for differentiation of Lycium ruthenicum explants was low.The optimum hormone combinations for differentiation of adventitious buds from hypocotyl,stem and leaf were 0.050 mg·L-16-BA+0.020 mg·L-1 NAA and 0.100 mg·L-16-BA+NAA 0.005 mg·L-1 and 0.200 mg·L-16-BA+0.005 mg·L-1 NAA,respectively.The proliferation numbers were 3 buds per segment,4 buds per segment and 5 buds per leaf.All buds could well root and grew in MS medium.With the optimum plant hormone combination,the adventitious buds could be induced from the upper,middle and lower stem segments and leaves of annual Lycium ruthenicum.The number of adventitious buds induced from the upper stem segments and leaves was significantly higher than that from the lower parts.
