基于挥发油出油量的逍遥片提取工艺优化及挥发性成分研究 被引量:5

摘要 目的优化水蒸馏法提取逍遥片中当归、白术挥发油的提取工艺,研究不同提取温度下挥发性成分的变化规律。方法采用析因实验设计筛选和优化提取工艺条件,利用气相色谱—质谱联用技术(GC-MS)对不同温度下提取挥发油中挥发性成分进行分离鉴定,峰面积归一化法计算各成分的相对百分含量。结果确定最佳提取工艺为加水量6倍水,蒸馏时间6 h,暴沸提取。在160℃提取下出油量最高,低温120℃主要提取挥发油中单萜烯类成分;130℃及以上主要为内酯类和单萜烯类成分。结论该提取工艺条件稳定可行,提取温度对挥发油中挥发性成分有一定影响,为以挥发性有效成分指导的逍遥片及其他大复方中药的开发利用提供参考依据及研究思路。 Objective To optimize the steam distillation process for extraction of volatile oil from Angelica sinensis(Oliv.)Diels and Atractylodes macrocephala Koidz.in Xiaoyao tablet and study the variation of volatile components at different temperatures.Methods The extraction process conditions are screened and optimized by the factorial experimental design.The volatile components extracted at different temperatures are separated and analyzed by the gas chromatographic-mass spectrometry(GC-MS),their relative percentage are calculated by the chromatographic peak area normalization method.Results The optimal extraction process is determined to be 6 times water addition,distillation time 6 h,and bump extraction.The highest oil volume is obtained at 160°C.The volatile components are mainly monoterpenes collected at 120°C,and the lactones and monoterpenes collected at 130°C and above.Conclusion The extraction process conditions are stable and feasible,and the the volatile components in the volatile oil are influenced by extraction temperature.It provides reference and research ideas for the development and utilization of Xiaoyao tablet and other complex Chinese medicines formula guided by volatile active ingredients.
