摘要
目的通过响应面法优化天麻抗氧化成分回流提取工艺,并研究其体外抗氧化活性。方法选择料液比、提取温度、提取时间为考察因素,羟基自由基清除率为评价指标,采用Box-Behnken响应面法优化提取工艺。通过羟基自由基清除率和DNA氧化损伤保护作用评价天麻体外抗氧化活性。结果天麻抗氧化成分回流提取最佳工艺为:料液比1∶55(g/ml),提取时间66 min,提取温度65℃。羟基自由基清除能力较好,IC50值为13.018 mg/ml。浓度为0.15 g/ml时具有最佳的DNA氧化损伤保护作用。结论该方法稳定可行,可用于提取天麻抗氧化成分,并且该成分具有一定的抗氧化活性。
objective To optimize the reflux extraction process of antioxidant components from Gastrodia elata by response surface method,and study its antioxidant activity in vitro.Methods The material-liquid ratio,extraction temperature and extraction time are selected as the investigation factors,hydroxyl radical scavenging rate is evaluation index,Box-Behnken response surface method is used to optimize the extraction process.The antioxidant activity of Gastrodia elata in vitro is evaluated by hydroxyl radical scavenging rate and DNA oxidative damage protection.Result The optimum reflux extraction technology of antioxidant components from Gastrodia elata is as follows:ratio of material to liquid is 1∶55(g/ml),extraction time is 66 min,extraction temperature is 65℃.The scavenging capacity of hydroxyl radicals is good,the IC50 value is 13.018 mg/ml.The best protective effect of DNA oxidative damage is obtained when the concentration is 0.15 g/ml.Conclusion The technique is stable and feasible,which can be used for the extraction of antioxidant components.
